摘要
目的证实胰岛素样生长因子结合蛋白5 (IGFBP-5)可以通过调节胸主动脉血管平滑肌细胞(VSMCs)的过度增殖影响高血压的发病。方法采用Western blot方法检测自发性高血压组大鼠(SHR)和血压正常组大鼠(WKY)原代培养的VSMCs中IGFBP-5的表达。采用MTT方法检测用不同剂量IGFBP-5处理WKY组VSMCs时对该细胞增殖的影响。结果 SHR组和WKY组相比较,IGFBP-5的蛋白表达量显著增高;MTT结果显示,IGFBP-5具有促进WKY组VSMCs增殖的作用。结论 IGFBP-5能够促进VSMCs的增殖进而可能影响高血压的发生、发展。
Objective To investigate insulin like growth factor binding protein-5(IGFBP-5)how regulates excessive vascular smooth muscle cell proliferation so that to develop hypertension.Methods The expression of IGFBP-5 in smooth muscle cells(VSMCs)from spontaneously hypertensive rats(SHR)and Wistar Kyoto rats(WKY)is detected with Western blot.The VSMCs of WKY cells were exposured to IGFBP-5 dose dependently for 24 hrs,then cell proliferation was determined by using an MTT assay.Results The levels of IGFBP-5 protein was found to be higher in the VSMC from SHR than in those from WKY.MTT assays revealed that recombinant IGFBP-5 dose dependently up-regulated VSMC proliferation in WKY to levels that were observed in SHR.Conclusion IGFBP-5 is contribute to the development of hypertension via stimulates VSMCs proliferation.
