消退素D1对牙周炎大鼠龈沟液中炎症因子影响的研究

The effects of resolvin D1 on the inflammatory factors in rats with periodontitis

目的:研究消退素D1(Resolvin D1,Rv D1)在大鼠牙周炎的发生发展过程中,对相关炎症因子表达的影响。方法:选取30只雄性SD大鼠,采用在左右上颌第二磨牙绑线的方法构建大鼠牙周炎模型。建模成功后,将大鼠随机分成5组(n=6),A组为牙周炎组,B、C、D、E组分别给予1、10、50、100 ng/ml进行干预。干预前和干预后7 d检测大鼠牙周状况并采用ELISA方法检测龈沟液中的IL-1β、TNF-α、IL-6的水平。结果:用药7 d后牙周探诊深度(probing depth,PD)明显降低(P <0. 001),各浓度组龈沟液中IL-1β、TNF-α、IL-6均明显降低(P <0. 05),其中D组、E组TNF-α的表达与治疗前差异具有显著性(P <0. 001);组间比较,IL-1β、TNF-α、IL-6各浓度组间差别无统计学意义,但IL-6的表达随着Rv D1浓度的升高而降低,具有一定的Rv D1浓度依赖性。结论:Rv D1可降低大鼠牙周炎龈沟液中炎症因子IL-1β、TNF-α、IL-6的水平。

Objective:To explore the effects of Resolvin D1(RvD1)on the expression of inflammatory cytokines in the progress of periodontitis.Methods:30 male SD rats were used as the periodontitis models established by binding wire to maxillary second molars.The rats were randomly divided into 5 groups(n=6):control group(A)of periodontitis,1 ng/ml RvD1+periodontitis group(B),10 ng/ml RvD1+periodontitis group(C),50 ng/ml RvD1+periodontitis group(D)and 100 ng/ml RvD1+periodontitis group(E).Probing depth(PD)was measured and gingival crevicular fluid(GCF)was taken for the test of IL-1β,TNF-αand IL-6 before and 7 days after injection of RvD1 by ELISA.Results:The PD was decreased after treatment in all treated rats(P<0.001).After treatment the levels of IL-1βand IL-6 in GCF of RvD1 were lower than those in the control group(P<0.05),and the TNF-αlevels of group D and E were decreased(P<0.001),and lower than those in group A,B and C(P<0.05).However,there was no significant difference of IL-1β,TNF-αand IL-6 among the different RvD1 concentration groups.But the expression of IL-6 was decreased with the increase of RvD1 concentration.Conclusion:Resolvins can reduce the expression of IL-1β,TNF-αand IL-6 in GCF of rats with periodontitis.
