摘要
目的探讨2型糖尿病发生时体内主要的免疫脏器之一脾脏的代谢功能变化过程。方法 20只健康雄性SD大鼠随机分为正常对照组(n=9)和2型糖尿病组(n=11),采用链脲佐菌素诱导制备2型糖尿病大鼠模型,取其脾脏组织并低温提取甲醇水提取液,进行核磁共振检测,对脾脏组织代谢轮廓的变化进行分析。结果多元统计分析结果显示,与正常对照组比较,2型糖尿病组大鼠脾脏提取液中β-羟基异丁酸、牛磺酸、α-葡萄糖、脯氨酸、β-葡萄糖、胆碱含量明显升高,同时亮氨酸、异亮氨酸、缬氨酸、丙氨酸、赖氨酸、乙酸、谷氨酰胺、蛋氨酸、谷氨酸、肉碱、甲胺、天冬氨酸、肌酸、乙醇胺、酪氨酸、组氨酸,苏氨酸,苯丙氨酸含量明显下降,差异有统计学意义(P<0.05)。结论 2型糖尿病模型大鼠脾脏组织中多种代谢物成分发生了明显的变化,这些代谢物的变化机制及其参与的代谢通路可能与2型糖尿病发生、发展过程的免疫损伤密切相关。
Objective To investigate the metabolic changes in spleen tissue of type 2 diabetes(T2DM)rats.Methods Twenty male Sprague-Dawley(SD)rats were divided into two groups:the healthy control group and T2DM group.After rat models were established,blood glucose was measured and spleen tissues metabolites were analyzed by 1 H-NMR spectroscopy.The data were preceding multivariate data analysis for differentiating the potential biomarkers.Results Compared to the control group,the T2DM rat models have significantly increased inβ-hydroxyisobutyrate,taurine,scyllo-inositol,α-glucose,proline,β-glucose,while decreased in leucine,Isoleucine,valine,alanine,lysine,acetate,glutamine,methionine,glutamate,carnitine,methylamine,aspartate,creatine,ethanolamine,tyrosine,histidine,Threonine,phenylalanine.Conclusion There are significant changes in the various metabolites in the spleen tissues of T2DM rat models,and these metabolites are involved in the immune damage of the T2DM.)
