紫草三黄栓抗炎作用机制及急性毒性研究 被引量:1

摘要 目的探讨紫草三黄栓的抗炎作用机制并考察该药的急性毒性。方法选取50只SD大鼠,随机分为空白组(蒸馏水)、对照组(马应龙痔疮栓0.32g/kg体质量)及实验组(紫草三黄栓高、中、低剂量3.62、1.81g/kg、0.91g/kg体质量),建立角叉菜胶致大鼠足肿胀和气囊炎2种炎症模型,以大鼠炎性足组织中的PGE2、MDA、NO含量、蛋白含量、SOD活力、COX-1、COX-2的含量和气囊渗出液中白细胞个数为指标,考察紫草三黄栓的抗炎机制。选取50只昆明小鼠,随机分为实验组(19.1g生药/kg体质量)及空白组(蒸馏水),直肠连续给药2w,考察紫草三黄栓的急性毒性。结果与空白组比较,紫草三黄栓高、中、低剂量组炎性组织中的PGE2、MDA、NO、蛋白含量降低,SOD活性提高,大鼠气囊渗出液中白细胞数减少,血清中COX-1和COX-2的表达水平降低,差异有统计学意义(P <0.05);与空白组比较,急毒实验给药组大鼠体量变化差异无统计学意义(P>0.05)。结论紫草三黄栓抗炎作用的机制与抑制PGE2、NO、COX-1、COX-2等炎症介质合成,抑制脂质过氧化、提升自由基清除能力,减少白细胞渗出有关,且安全无毒。 Objective To explore the anti-inflammatory mechanism of Zi-Cao-San-Huang suppository and investigate the acute toxicity of the drug.Methods Fifty SD rats were randomly divided into blank group(distilled water),control group(Ma-Ying-Long hemorrhoid suppository 0.32 g/kg)and experimental group(Zi-Cao-San-Huang suppository in the high dose of 3.62 g/kg,medium dose of 1.81 g/kg,low dose of 0.91 g/kg),Two inflammatory models of rat foot swelling and airbag inflammation induced by carrageenin were established.The anti-inflammatory mechanism of Zi-Cao-San-Huang suppository was investigated by taking the content of PGE-2,MDA,NO,protein content,SOD activity,COX-1,COX-2 content and the number of white blood cells in the airbag exudate as indexes;50 Kunming mice were randomly divided into experimental group(19.1 g/kg)and blank group(distilled water),and the rectum was administered with the drug continuously for 2 w to observe the acute toxicity of the mice.Results Compared with the blank group,the contents of PGE-2,MDA,NO and protein in inflammatory tissue in the high,medium and low dose groups were significantly reduced,the activity of SOD was increased,the white blood cell number was reduced in the airbag exudate of rats and the expression levels of COX-1 and COX-2 in serum were inhibited,and the differences were statistically significant(P<0.05).Compared with the blank group,there was no significant difference in body weight(P>0.05).Conclusion The mechanism of the anti-inflammatory effect of Zi-Cao-San-Huang Suppository is related to inhibition of the synthesis of inflammatory mediators such as PGE-2,NO,COX-1 and COX-2,inhibition of lipid peroxidation,improvement of free radical scavenging ability,and reduction of the exudation of white blood cells.And it is non-toxic.
