Notch 2、Jagged-2在心肌梗死合并糖尿病大鼠心肌中表达研究

摘要 目的探讨Notch 2、Jagged-2在心肌梗死合并糖尿病大鼠心肌中表达。方法选取6~8周龄健康雄性SD大鼠30只,随机分为对照组、糖尿病(DM)组、糖尿病假手术(DM-sham)组、心肌梗死(MI)组、心肌梗死合并糖尿病(DM-MI)组,每组6只,建立模型。应用酶联免疫吸附法(ELISA)测定各实验组大鼠血清中磷酸肌酸激酶同功酶(CK-MB)和肌钙蛋白的水平,采用实时荧光定量聚合酶链式反应方法检测大鼠心肌中Notch 2和Jagged-2 m RNA的表达水平,采用Western blot方法检测各实验组大鼠心肌中Notch 2和Jagged-2蛋白的表达水平。结果 MI组、DM-MI组CK-MB浓度、肌钙蛋白浓度与对照组、DM组和DM-sham组比较,差异均有统计学意义(P <0. 05)。DM组、DM-sham组及MI组三组Notch 2 m RNA和蛋白表达水平均显著高于对照组,差异均有统计学意义(P <0. 05); DM-MI组与对照组、DM组、DM-sham组、MI组比较,Notch 2 m RNA和蛋白表达水平均降低,差异均有统计学意义(P <0. 05)。DM组、DM-sham组、MI组、DM-MI组Jagged-2 m RNA表达水平均显著高于对照组,差异均有统计学意义(P <0. 05); DM组、DM-sham组、MI组Jagged-2蛋白表达水平均显著高于对照组,差异均有统计学意义(P <0. 05)。结论在单纯糖尿病和单纯心肌梗死大鼠的心肌组织中,Notch受体和配体的表达均升高的趋势,而心肌梗死合并糖尿病大鼠的Notch信号通路各指标反而呈降低的趋势。
