摘要
以5个月季F1代AY02、AY03、YK01、CK01、CK04植株的带芽茎段为外植体,进行组培快繁研究,比较其在启动培养基上的萌芽率,筛选适宜的增殖培养基、生根培养基。结果表明,上述5个材料外植体在启动培养基MS+2 mg/L 6-BA+0. 1 mg/L NAA上的萌发率分别为64. 00%、72. 41%、83. 33%、77. 78%、76. 19%。AY02最适增殖培养基为MS+2 mg/L 6-BA+0. 1 mg/L NAA,最适生根培养基为1/2 MS+0. 1 mg/L NAA; AY03、YK01、CK01最适增殖培养基为MS+2 mg/L 6-BA+0. 05 mg/L NAA,最适生根培养基为1/2 MS+0. 2 mg/L NAA; CK04最适增殖培养基为MS+2 mg/L 6-BA+0. 05 mg/L NAA和MS+2 mg/L 6-BA+0. 1 mg/L NAA,最适生根培养基为1/2 MS+0. 2 mg/L NAA。
Stems contained axillar buds of five F 1 rose AY02,AY03,YK01,CK01,CK04 were used as explants for tissue culture experiment.The germination rates of five F 1 rose on initiation medium were compared.The optimum proliferation medium and rooting medium of five F 1 rose were selected.The results showed that the germination rate of five above-mentioned materials on initiation medium(MS+2 mg/L 6-BA+0.1 mg/L NAA)was 64.00%,72.41%,83.33%,77.78%and 76.19%,respectively.The optimum proliferation medium for AY02 was MS+2 mg/L 6-BA+0.1 mg/L NAA,the optimum rooting medium for AY02 was 1/2 MS+0.1 mg/L NAA;The optimum proliferation medium for AY03,YK01 and CK01 was MS+2 mg/L 6-BA+0.05 mg/L NAA,the optimum rooting medium for AY03,YK01 and CK01 was 1/2 MS+0.2 mg/L NAA;The optimum proliferation media for CK04 were MS+2 mg/L 6-BA+0.05 mg/L NAA and MS+2 mg/L 6-BA+0.1 mg/L NAA,the optimum rooting medium for CK04 was 1/2 MS+0.2 mg/L NAA.
