摘要
制备了一种基于二茂铁增强ABEI的电化学发光测定8-OH-dG的电化学适配体传感器.8-OH-dG的适配体一部分与固定在金电极上的cDNA互补配对,另一部分与末端修饰二茂铁的pDNA互补配对.由于二茂铁的引入,ABEI的电化学发光信号将显著增强.然而,当8-OHdG存在时,其可以诱导富G序列适配体形成紧密的G-四倍体结构,电极表面引入的二茂铁数量减少,导致ABEI的电化学发光信号降低.在最佳实验条件下,ABEI发光强度与8-OH-dG浓度的对数呈线性关系,线性范围为1nM-100μM,检出限为0.33nM.已将该方法应用于人体尿液中8-OH-dG的检测,结果满意,因此该方法在临床检测中具有潜在的应用价值.
In this work,an aptasensor was prepared for the determination of 8-OH-dG based on ferrocenescence enhanced ABEI electrochemical luminescence.The aptamer of 8-OH-dG is partially complementary paired with the capture DNA immobilized on the gold electrode,and the other part is complementary paired with the p DNA of the terminally modified ferrocenescence.Due to the introduction of ferrocenescence,the electrochemiluminescence signal of ABEI will be significantly enhanced.However,in the presence of 8-OH-dG,the conformation of Apt changed from single-stranded structure to tight G-quadruplex structure,and the number of ferrocenescence introduced on the surface of the electrode decreases,the electrochemi luminescence signal of ABEI also decrease.Under the optimal experimental conditions,the logarithm of ABEI luminescence intensity was linear with the logarithm of 8-OH-dG concentration in the range from 1nM to 100μM with the detection limit of 0.33 nM.This method is applied to the detection of 8-OH-dG in human urine with satisfactory results,so this method has potential application value in clinical detection.
