摘要
为探究鸡血藤多糖的理化性质及活性,将经过DEAE-52纤维素离子交换柱分离纯化得到的鸡血藤多糖(MDP1)采用氯磺酸—吡啶法进行硫酸化修饰,得到硫酸化修饰的多糖(S-MDP1),测得其取代度为1. 94。采用TGA和DSC对两种多糖的热性质进行分析,结果表明,两种多糖均具有良好的热稳定性能。通过羟基自由基清除实验对两种多糖的体外抗氧化活性进行了评价,结果表明,在测试的浓度范围内(0. 2~2. 0 mg/m L)这两种多糖对羟基自由基均有一定的清除能力,并表现出剂量依赖性。采用MTT比色法测定了两种多糖在体外对小鼠巨噬细胞增殖的影响,测试结果表明,MDP1和S-MDP1在50~200μg/m L的浓度范围内均能促进小鼠巨噬细胞的增殖,可见MDP1和S-MDP1可以作为潜在的抗氧化剂及免疫调节剂。
To study the physicochemical properties and activity of polysaccharides from Millettia dielsiana,the polysaccharide(MDP1)was isolated and purified by DEAE-52 cellulose column and then sulfated with chlorosulfonic acid-pyridine method to obtain the sulfated polysaccharide(S-MDP1).The degree of substitution is 1.94 of S-MDP1.The thermal properties of the two polysaccharides were analyzed by TGA and DSC,and the results show that the two polysaccharides had good thermal stability properties.The antioxidant activities of the two polysaccharides were evaluated by hydroxyl radical scavenging assay,and the results show that both polysaccharides have scavenging ability for hydroxyl radicals in the tested concentration range(0.2~2.0 mg/mL)and exhibited a dose-dependent manner.The effects of both polysaccharides on the proliferation of macrophages were measured by MTT assay in vitro,and the results show that MDP1 and S-MDP1 could significantly promote the proliferation of macrophages in the concentration range of 50~200μg/mL.These results imply that MDP1 and S-MDP1 had the potential to be used as antioxidant and immunomodulatory.
