HPLC-ELSD法同时测定益肺清化颗粒中的黄芪甲苷、桔梗皂苷D和麦冬皂苷D的含量

Determination of astragaloside Ⅳ,platycodin D,ophiopogonin D in Yifeiqinghua granules by HPLC-ELSD

目的:建立益肺清化颗粒中黄芪甲苷、桔梗皂苷D、麦冬皂苷D的定量测定方法。方法:采用高效液相色谱-蒸发光散射检测(HPLC-ELSD)法,色谱柱为SB-C_(18)(250 mm×4. 6 mm,5μm),流动相为乙腈-水(20∶80);体积流量1. 0 ml/min;柱温30℃; ELSD漂移管温度95℃,气体流量2. 5 L/min。结果:黄芪甲苷、桔梗皂苷D、麦冬皂苷D的线性范围分别为1. 5～30μg/ml(r=0. 999 2)、1. 0～20μg/ml(r=0. 999 3)和0. 5～10μg/ml(r=0. 999 2),回收率分别为99. 59%(RSD为1. 39%,n=9)、99. 88%(RSD为0. 68%,n=9)和100. 16%(RSD为2. 39%,n=9)。结论:该方法测定简便、快速、重复性好,可作为益肺清化颗粒的质量控制方法。

Objective:To establish a method for the quantitative determination of astragalosideⅣ,platycodin D,ophiopogonin D in Yifeipinghua granules.Methods:HPLC-ELSD was used.The analysis was carried on SB-C 18(250 mm×4.6 mm,5μm)column with mobile phase consisting of acetoneitrile and water in a ratio of 20(acetoneitrile)to 80(water).The flow rate was 1.0 ml/min.The column temperature was 30℃.The drift tube temperature was 95℃.The flow rate of gas was 2.5 L/min.Results:The responses of astragalosideⅣ,platycodin D,ophiopogonin D were liner in the ranges of 1.5~30μg/ml(r=0.999 2),1.0~20μg/ml(r=0.999 3),and 0.5~10μg/ml(r=0.999 2),respectively.The average recoveries were 99.59%(RSD=1.39%,n=9),99.88%(RSD=0.68%,n=9),and 100.16%(RSD=2.39%,n=9),respectively.Conclusion:The method is simple,rapid,and with good repeatability.It can be used for the quality control of Yifeiqinghua granules.
