摘要
目的探讨大鼠骨髓间充质干细胞(BMSCs)向血管平滑肌细胞(VSMCs)分化过程中小窝蛋白-1(caveolin-1)的表达变化。方法采用全骨髓培养法,体外分离培养SD大鼠BMSCs,成骨成脂成软骨检测干细胞的分化潜能,流式细胞仪鉴定干细胞表面标志物。体外TGF-β1诱导BMSCs向VSMCs分化,Western blot检测SM-MHC、SM22α、caveolin-1和myocardin的表达。结果 BMSCs在TGF-β1诱导下可分化为VSMCs,诱导培养组细胞caveolin-1、SM-MHC及myocardin表达的水平随诱导时间的增加而增强(P<0. 05)。结论 BMSCs体外诱导可以分化为VSMCs,小窝蛋白-1与干细胞来源的VSMCs的形成有关。
Objective To explore the expression of caveolin-1 in the differentiation of rat bone marrow mesenchymal stem cells(BMSCs)into vascular smooth muscle cells(VSMCs).Methods BMSCs from SD rats were cultured by whole-bone marrow culture.Then,testing the differentiation potential of stem cells through adipogenic,osteoplastic and chondrogenic differentiation.BMSCs were induced to differentiate into VSMCs by transforming growth factor-β1(TGF-β1)in vitro.Western blot was performed to evaluate the expression of SM-MHC,SM22αand caveolin-1.Results TGF-β1 treatment increased the expression of caveolin-1,SM-MHC and myocardin in a time dependent manner.During the stage of BMSCs differentiate into VSMCs,the expression of caveolin-1 was specifically increased(P<0.05).Conclusions Caveolin-1 may be related to the formation of smooth muscle cells derived from stem cells.
