不同剂量异鼠李素对炎症损伤小鼠巨噬细胞RAW264.7增殖能力的影响及其机制探讨 被引量:7

摘要 目的观察异鼠李素(ISO)对炎症损伤小鼠巨噬细胞RAW264. 7增殖能力的影响,并探讨其机制。方法取对数生长期RAW264. 7细胞,将细胞分为5组,低、中、高剂量组分别加入已经配好含10 ng/m L脂多糖(LPS)及低、中、高剂量ISO(50、100、200μg/m L)的溶液,模型组加入含10μg/m L LPS的1640培养基(含10%FBS),对照组加入含10%溶剂对照的1640完全培养基。测算各组细胞增殖指数,检测各组一氧化氮(NO)、肿瘤坏死因子(TNF-α)。结果与对照组比较,模型组细胞12、24、36、48 h细胞增殖指数增加(P均<0. 05);与模型组比较,低剂量组24、36 h及中剂量组和高剂量组12、24、36、48 h细胞增殖指数减少(P均<0. 05);与低剂量组比较,中剂量组24 h及高剂量组24、48 h细胞增殖指数减少(P均<0. 05)。与对照组比较,模型组NO、TNF-α水平升高(P均<0. 05);与模型组比较,低剂量组NO水平及中剂量组和高剂量组NO、TNF-α水平降低(P均<0. 05);与低剂量组比较,中剂量组和高剂量组NO、TNF-α水平降低(P均<0. 05)。结论 ISO可抑制RAW 264. 7细胞增殖能力,且呈剂量依赖性,高剂量作用最大;机制可能与抑制NO、TNF-α有关。 Objective To observe the effect of isorhamnetin(ISO)on the proliferation of macrophage RAW264.7 in mice with inflammatory injury and to explore its mechanism.Methods RAW264.7 cells in the logarithmic growth phase were divided into 5 groups:the low-,medium-and high-dose groups which were added with 10 ng/mL lipopolysaccharide(LPS)and low-,medium-and high-dose ISO solution(50,100,and 200μg/mL),the model group which was added with 10μg/mL LPS in 1640 medium(containing 10%FBS),and the control group which was added with 1640 complete medium containing 10%solvent.The cell proliferation index of each group was measured,and nitric oxide(NO)and tumor necrosis factor-α(TNF-α)were detected in each group.Results Compared with the control group,the cell proliferation index of the model group increased at 12,24,36,48 h(P<0.05);compared with the model group,the cell proliferation index decreased in the low-dose group at 24 and 36 h,and the medium-dose group and the high-dose group at 12,24,36 and 48 h(P<0.05).Compared with the low-dose group,the cell proliferation index decreased in the medium-dose group at 24 h and the high-dose group at 24 and 48 h(P<0.05).Compared with the control group,the levels of NO and TNF-αin the model group increased(both P<0.05).Compared with the model group,the NO level in the low-dose group,and the NO and TNF-αlevels in the medium-dose group and the high-dose group decreased(all P<0.05);compared with the low-dose group,the levels of NO and TNF-αdecreased in the medium-dose group and the high-dose group(both P<0.05).Conclusion ISO can inhibit the proliferation of RAW 264.7 cells in a dose-dependent manner by inhibiting the NO and TNF-αlevels,and it has the greatest effect at the highest dose.
