摘要
目的:通过检测下肢动脉粥样硬化合并血栓形成患者与正常人血小板中微小RNAs(mi RNAs)的差异表达情况,探究异常表达的mi RNAs参与调节血小板功能可能的通路。方法:应用微阵列技术对16例动脉粥样硬化合并血栓和4例正常人血小板进行mi RNAs分析。对显著差异表达的mi RNA对应的靶基因以及所有靶基因进行GO与Pathway富集分析。结果:通过micro RNAs的基因芯片分析,共发现299个表达上调的micro RNAs,102个表达下调的micro RNAs。筛选20个表达显著升高的基因和20个表达显著降低的基因进行GO与Pathway富集分析发现,主要与凋亡过程的负调节、MAPK介导的信号转导通路相关。结论:mi RNA可能通过凋亡过程的负调节、MAPK介导的信号转导通路相关通路参与了血小板功能的调控,从而影响动脉粥样硬化患者血栓形成。
Objective:To investigate the molecular mechanism of abnormal expression of microRNAs in platelets of patients with atherothrombosis of lower extremity and normal subjects,and explore the possible pathway of abnormal expression of miRNAs in regulating platelet function.Methods:Microarray technique was used to analyze the microRNAs of platelets in 16 atherosclerotic patients with thrombosis and 4 normal subjects.GO and Pathway enrichment analysis were performed on the target genes and all target genes of the differentially expressed microRNAs.Results:A total of 299 up-regulated microRNAs and 102 down-regulated microRNAs were found by microRNAs microarray analysis.Twenty genes with significantly increased expression and 20 genes with significantly decreased expression were screened for GO and Pathway enrichment analysis.The results showed that GO and Pathway enrichment were mainly related to the negative regulation of apoptosis and MAPK-mediated signal transduction pathway.Conclusion:miRNA may be involved in the regulation of platelet function through the negative regulation of apoptosis and MAPK-mediated signal transduction pathway,which may affect the atherothrombosis patients.
