摘要
目的:探讨长链非编码(lncRNA)ANRIL靶向miR-214的表达调控卵巢癌细胞增殖与侵袭行为及其机制。方法:qPCR检测ANRIL和miR-214在卵巢癌组织中的表达情况及ANRIL在不同卵巢癌细胞株中的表达水平;分析ANRIL和卵巢癌患者的临床病理资料之间的关联;双荧光素酶报告基因检测ANRIL与miR-214之间的相互作用;克隆形成实验和MTT增殖实验检测抑制ANRIL的表达后卵巢癌细胞的增殖能力的变化情况,Transwell侵袭实验检测抑制ANRIL的表达后卵巢癌细胞的侵袭能力的变化情况;裸鼠体内成瘤实验ANRIL对卵巢癌细胞成瘤能力的影响。结果:与正常卵巢组织相比,ANRIL和miR-214在卵巢癌组织中的表达相对较高,与其他卵巢癌细胞株相比,SKOV3细胞中ANRIL表达最高,miR-214的表达水平最高;ANRIL的表达与卵巢癌的病理分期相关以及淋巴结转移情况有关,随着分期越高,ANRIL在卵巢癌组织中表达越高,淋巴结转移的患者中ANRIL的表达也相对较高;双荧光素酶实验证实ANRIL能与miR-214的3′UTR特异性结合,调控miR-214的表达与活性;抑制ANRIL的表达后可以促进卵巢癌细胞的增殖和侵袭能力;抑制ANRIL的表达后,裸鼠体内成瘤能力受到一定的抑制。结论:ANRIL可以调控miR-214的表达,影响卵巢癌细胞的增殖和侵袭能力。
Objective:To investigate the effect of long-chain non-coding(lncRNA)ANRIL targeting miR-214 on the proliferation and invasion of ovarian cancer cells and its mechanism.Methods:The expression of ANRIL and miR-214 in different ovarian cancer cell lines was detected by qPCR.The association between ANRIL and clinicopathological data in patients with ovarian cancer was analyzed.Double luciferase reporter gene was used to detect the interaction between ANRIL and miR-214.Clone formation assay and MTT proliferation assay were used to detect the proliferation of ovarian cancer cells after ANRIL inhibition.Transwell invasion assay was used to detect the invasion ability of ovarian cancer cells after ANRIL inhibition.Effect of ANRIL on tumorigenesis of ovarian cancer cells in nude mice was detected.Results:Compared with other ovarian cancer cell lines,the expression of ANRIL was the highest in SKOV3 cells and the expression level of miR-214 was the highest.The expression of ANRIL was correlated with the pathological stage of ovarian cancer and the lymph node metastasis.The higher the expression of ANRIL in ovarian cancer tissues,the higher the expression of ANRIL in patients with lymph node metastasis.Double luciferase experiments confirmed that ANRIL could bind specifically to the 3′UTR of miR-214.Inhibition of ANRIL expression could promote ovarian cancer cell proliferation and invasion ability.Inhibition of ANRIL expression inhibited tumorigenic ability of nude mice in vivo to be a certain extent.Conclusion:ANRIL can regulate the expression of miR-214 and affect the proliferation and invasion of ovarian cancer cells.
