摘要
目的:探讨孕期微波辐射暴露对仔鼠脑前额叶皮质5-羟色胺2A受体(5-HT_(2A)R)表达的影响。方法:SD孕鼠随机分为空白对照组(常规饲养)、低剂量辐射组(全孕期,6h/d,)和高剂量辐射组(全孕期,24h/d);各组仔鼠在出生后第1、7、30、60天取脑前额叶皮质,采用免疫组织化学和Real-time荧光PCR法检测5-HT_(2A)R mRNA的表达。结果:免疫组织化学结果显示5-HT_(2A)R表达在前额叶皮质神经元上,与空白对照组比较,2个辐射组的5-HT_(2A)R表达水平均显著降低,且高剂量辐射组显著低于低剂量辐射组;Real-time荧光PCR法检测结果与免疫组织化学结果一致。结论:孕期微波辐射暴露可下调仔鼠脑前额叶皮质5-HT_(2A)R的表达。
Objective:To determine the effects of microwave radiation exposure during pregnancy on 5-HT 2A receptor expression in the prefrontal cortex of offspring rats.Methods:Twenty-four SD pregnant rats were randomly divided into blank control group(conventional breeding),low-dose radiation group(during the whole pregnancy,six hours per day)and high-dose radiation group(during the whole pregnancy,twenty-four hours per day).It was randomized to keep sixty-four offspring rats respectively in each group,and the prefrontal cortex of the brain was taken 1 day,7 days,30 days and 60 days after birth.Immunochemical and real-time fluorescence PCR methods were used to measure the expression of 5-HT 2A receptor in the prefrontal cortex.Results:The results of the immunochemical method indicated that 5-HT 2A receptor was expressed in the neurons,and the expression of 5-HT 2A receptor in the two radiation groups were significantly lower than that in the blank group.Compared to the low-dose radiation group,the expression of 5-HT 2A receptor in the high-dose radiation group was significantly lower.The results of the real-time fluorescence PCR were consistent with the results of the immunochemical method.Conclusion:Microwave radiation exposure during pregnancy can down-regulate the expression of 5-HT 2A receptor in the prefrontal cortex of offspring rats.
