摘要
目的优化伤科解毒颗粒最优提取工艺。方法以干膏质量分数和绿原酸质量分数为考察指标,对试验结果进行加权综合评分,其中干膏权重系数为0.7,绿原酸权重系数为0.3。通过考察加水量(A)、浸泡时间(B)、煎煮时间(C)、煎煮次数(D)四个单因素,确定正交试验水平,筛选最佳提取工艺,并进行提取工艺验证。结果伤科解毒颗粒的最佳提取工艺为A_2B_3C_2D_2,即加24倍水,浸泡45 min,煎煮2次,每次1 h。提取验证该工艺的干膏质量分数和绿原酸质量分数的平均值为0.31 g/g和5.44 mg/g,RSD分别为1.9%和3.8%。结论该工艺简单稳定可行,可适用于后续的实验研究及工业大生产。
Objective To optimize the optimal extraction technology of Shangkejiedu Granules.Methods The dry extract mass fraction and chlorogenic acid mass fraction were taken as investigation indicators,and the test results were weighted comprehensive scores,among which the dry extract weight coefficient was 0.7,and the chlorogenic acid weight coefficient was 0.3.By investigating four single factors of water addition amount(A),soaking time(B),decoction time(C)and decoction times(D),the orthogonal test level was determined,the best extraction technology was screened,and the extraction technology was verified.Results The best extraction technology of Shangkejiedu Granules was A2B3C2D2,that was adding 24 times water,soaking for 45 minutes,decocting twice for 1 hour each time.The average values of dry extract mass fraction and chlorogenic acid mass fraction were 0.31 g/g and 5.44 mg/g,RSD was 1.9%and 3.8%respectively.Conclusion This technology is simple,stable and feasible,and can be applied to subsequent experimental research and industrial production.
