摘要
目的观察大鼠经锂—匹罗卡品(LI-PLO)诱发癫痫持续状态(SE)性脑水肿后水通道蛋白4(AQP4)的表达及其与癫痫持续状态性脑水肿的关系。方法实验于2017年4—10月在河北省人民医院实验中心完成。正常对照组(n=24)腹腔注射生理盐水,SE组(n=120)按癫痫发作后6 h、12 h、24 h、3 d、7 d分为5个亚组(n=24),SE组采用LI-PLO建立大鼠癫痫持续状态(SE)模型,造模成功后在不同时间点取材,干湿重法检测脑组织含水量。HE染色进行细胞形态学观察;免疫组织化学染色后用病理图文分析系统半定量检测APQ4的表达; RT-PCR检测AQP4mRNA表达。结果与正常对照组比较,SE组癫痫持续状态后6 h脑含水量和大脑组织AQP4表达、AQP4 mRNA表达均增加,24 h达高峰,持续3 d后下降,7 d后接近正常,但差异均有统计学意义(P <0. 05)。AQP4表达的平均光密度(OD)值与脑含水量呈显著正相关(r=0. 882,P=0. 005)。结论 AQP4表达上调与癫痫持续状态性脑水肿的形成与发展密切相关。
Objective To investigate the expression of aquaporin 4(AQP4)after status epilepticus(SE)induced by LiCl-Pilocarpine(LI-PLO)in rats,and the correlation with brain edema following SE.Methods SE models were established by LI-PLO,each cerebral tissue was respectively acquired at the designated time point.Pathological changes were observed through light microscope after HE staining;the expression of APQ4 was detected semi-quantitatively by immunohistochemical staining;AQP4 mRNA expression was measured by RT-PCR;Brain water content was determined according to the wet-dry weight method.Results The brain water content and expression of AQP4 began to increase at 6h after SE,reached their peaks at 24h,and continued to 3 d.Then they started to decrease and approached to the normal level at 7d(P<0.05).The average optical density(OD)of AQP4 expression and brain water content was significantly positively related(r=0.882,P=0.005).Conclusion The up-regulation of AQP4 expression was closely related with brain edema formation and development after SE.
