Simultaneous determination of six flavonoids in Naoxinqing Tablets by QAMS

摘要 目的建立一测多评法同时测定脑心清片(柿叶提取物)中金丝桃苷、异槲皮苷、槲皮苷、杨梅素、槲皮素、山柰酚的含有量。方法该药物甲醇提取液的分析采用Agilent Eclipse Plus C18色谱柱(250 mm×4. 6 mm,5μm);流动相乙腈-0. 1%磷酸,梯度洗脱;体积流量1. 0 m L/min;检测波长360 nm;柱温30℃。以槲皮素为内标,计算金丝桃苷、异槲皮苷、槲皮苷、杨梅素、山柰酚的相对校正因子,并测定其含有量。结果 6种黄酮在各自范围内线性关系良好(r≥0. 999 4),平均加样回收率99. 59%~101. 16%,RSD 1. 28%~2. 54%,一测多评法所得结果与外标法接近。结论该方法简便、准确、可靠,可用于脑心清片的质量控制。 AIM To establish a quantitative analysis of multi-components by single-marker(QAMS)method for the simultaneous content determination of hyperoside,isoquercitrin,quercitrin,myricetin,quercetin and kaempferol in Naoxinqing Tablets(extract from the leaves of Diospyros kaki Thunb.).METHODS The analysis of methanol extract of this drug was performed on a 30℃thermostatic Agilent Eclipse Plus C8 column(250 mmX4.6 mm,5"m),witli the mobile phase comprising of acetonitrile-0.1%phosphoric acid flowing at 1.0 mMmin in a gradient elution manner,and thedetection wavelength was set at 360 nm.With quercetin as an internal standard,the relative correction factors of hyperoside,isoquercitrin,quercitrin,myricetin and kaempferol were calculated,after which the content determination was made.RESULTS Six flavonoids showed good linear relationships with-in their own ranges(r)0.999 4),whoeaverage recoveries were 99.59%-101.16%with theRSDs of 1.28%-2.54%.The results obtained by QAMS approximated those obtained by external standard method.CONCLUSION This simple,accurate and reliable method can be used for the quality control of Naoxinqing Tablets.
