摘要
目的检测Claudin-1蛋白在食管癌细胞中的表达,并探讨其对食管鳞癌癌细胞的的自噬的影响。方法采用western blot检测Claudin-1在食管癌各细胞株的表达情况。SiRNA干扰Claudin-1蛋白的表达后,western blot检测干扰效果。mRFP-GFP-LC3腺病毒转染SiR组及NC组后,用激光共聚焦显微镜分析siR组与NC组的细胞自噬情况。结果细胞株KYSE 150的Claudin-1蛋白表达量最高,Si-RNA干扰Claudin-1的表达后siR组的自噬水平显著高于NC组(P<0. 05)。结论 Claudin-1蛋白的表达增多对食管癌细胞的细胞自噬有促进作用。
Objective To detect the expression of Claudin-1 protein in esophageal carcinoma cells and to investigate its effect on autophagy of esophageal squamous cell carcinoma cells.Methods Western blot was used to detect the expression of Claudin-1 in esophageal carcinoma cell lines.After SiRNA interfered with the expression of Claudin-1 protein,western blot was used to detect the interference effect.After mRFP-GFP-LC3 adenovirus was transfected into SiR group and NC group,the autophagy of siR group and NC group was analyzed by confocal laser microscopy.Results The expression level of Claudin-1 protein in cell line KYSE 150 was the highest.The level of autophagy in siR group was significantly higher than that in NC group after Si-RNA interfered with Claudin-1 expression(P<0.05).Conclusion The increased expression of Claudin-1 protein promotes the autophagy of esophageal cancer cells.
