白藜芦醇联合电针治疗运动性骨骼肌肉损伤的实验研究 被引量:5

摘要 目的:探讨电针(EA)联合白藜芦醇(Res)对运动性骨骼肌损伤(EIMD)的治疗效果。方法:取雄性SD健康大鼠50只,随机分为正常对照组(A组),EIMD组(B组)、Res+EA+EIMD组(C组)、Res+EIMD组(D组)和EA+EIMD组(E组)。运动后7d,取腹主动脉血检测丙二醛、超氧化物歧化酶、肌酸激酶、乳酸脱氢酶、白细胞介素-6和肌红蛋白的含量,并通过电镜观察骨骼肌超微结构变化以及采用western-blot技术检测腓肠肌中细胞膜骨架蛋白dystrophin以及热休克蛋白(HSP)-70的表达变化。结果:B组大鼠骨骼肌超微结构发生损伤,且各项血液生化指标和HSP-70表达量比A组升高,而dystrophin表达量降低。C组、D组和E组大鼠骨骼肌超微结构变化较B组轻,各项血液生化指标均低于B组,HSP-70和dystrophin的表达量显著高于B组,且C组的治疗效果明显优于D组和E组(P <0. 05)。结论:白藜芦醇和电针具有促进EIMD修复的功效,两者相结合可使疗效更显著,具有明显协同增效作用。 Objective:To observe the effect of combined application of Resveratrol(Res)and electroacupuncture(EA)in exercise induced muscle damage(EIMD)in rats.Methods:Fifty male SD rats were divided randomly into five groups:normal control group(group A,n=10),EIMD group(group B,n=10),EIMD+EA+Res group(group C,n=10),EIMD+Res group(group D,n=10)and EIMD+EA group(groupE,n=10).The blood samples were drawn from aorta abdominalis and malonaldehyde,superoxide dismutase,creatine kinase,lactic dehydrogenase,interleukina-6 and myohemoglobin were detected with biotechnology.Histological changes of muscle fiber in gastrocnemius muscle were observed under transmission electron microscope(TEM),and the expression of dystrophin and HSP-70 were detected with western-blot assay.Results:In group B,the ultramicrostructure of gastrocnemius muscles have damaged,and the level of biochemical indicators in blood and expression of dystrophin in muscle were higher,and expression of HSP-70 in muscle was lower than that in group A.The level of biochemical indicators in blood declined obviously in group C,D and E,and the ultrastructure of skeletal muscle in group C,D and E changed less obviously,the expression of dystrophin and HSP-70 were higher than that in group B.Furthermore,in group C,the repair effect of ultrastructure of skeletal muscle was batter,the level of these indicators in blood were lower and the expression of dystrophin and HSP-70 were higher than that in group D and E.Conclusion Both Res and EA could promote the reparation of skeletal muscle after EIMD,and the combined application could provides superior clinical effectiveness.
