安徽和县地区辣椒病株辣椒轻斑驳病毒的鉴定

Identification of Pepper Mild Mottle Virus in Hexian,Anhui Province

[目的]鉴定安徽和县地区辣椒病株辣椒轻斑驳病毒(pepper mild mottle virus,PMMoV)(PMMoV-AH)。[方法]采用胶体金免疫层析试纸条和RT-PCR检测和县辣椒病样辣椒轻斑驳病毒,并将其外壳蛋白基因序列与已报道的国内外12个PMMoV分离物进行同源性分析。[结果]PMMoV-AH与已报道的12个分离物核苷酸同源性介于94.5%～100.0%,氨基酸同源性介于96.8%～100.0%。基于外壳蛋白基因序列进行系统发育分析发现,PMMoV-AH与亚洲分离物亲缘关系密切,与其他区域分离物亲缘关系较远。[结论]PMMoV危害性强,对于其在安徽地区的发生和防治还需进一步研究。

[Objective]To identify the pepper mild mottle virus in Hexian,Anhui Province.[Method]The PMMoV-AH was identified in the sample of diseased fruits of pepper which collected in Hexian by using immunochromatography colloidal gold strip and RT-PCR method.The pair of universal primers was used to amplify the coat protein gene sequence of PMMoV-AH.[Result]Sequence analysis and phylogenetic analysis showed that PMMoV-AH was closely related to 12 PMMoV isolates reported in China and abroad with nucleotide sequence identity between 94.5%-100.0%and amino acid sequence identity between 96.8%-100.0%.Phylogenetic tree showed that PMMoV-AH was closely related to Asian isolates but not with other isolates.[Conclusion]Due to the significant losses caused by PMMoV,further research is required to control and prevent the prevailing of the disease caused by PMMoV in Anhui Province.
