原花青素和乌索酸对小鼠酒精性肝损伤保护作用的比较研究

摘要 目的研究原花青素和乌索酸两种生物活性物质对酒精诱发的小鼠肝损伤的保护功能。方法将同批小鼠饲养建立酒精性肝损伤模型后,随机为4组,分别使用原花青素组和乌索酸对2组保护组小鼠进行灌胃,灌胃2周后,眼球取血统一处死;收集其肝脏,制作肝匀浆与切片,再使用试剂盒测试其血清肝匀浆的超氧化物歧化酶(SOD)、丙二醛(MDA)、天冬氨酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT)等指标,并通过切片观察不同组小鼠肝损伤情况。结果对4组小鼠在灌胃期间的体重对比发现,不同时间小鼠体重的变化差异明显,第7天和第14天体重变化差异有统计学意义(P <0. 05);乌索酸组和原花青素组小鼠不同天数体重比较差异有统计学意义(P <0. 05);对4组小鼠各项指标比较,其中AST(F=9. 836,P=0. 000)、ALT(F=7. 539,P=0. 000)、MDA(F=28. 829,P=0. 000)、SOD(F=8. 491,P=0. 000),差异均有统计学意义;原花青素和乌索酸小组小鼠各指标比较,差异均有统计学意义(P <0. 05);切片观察发现乌索酸组损伤较原青花素组更为严重。结论酒精组小鼠其肝脏的各项指标变化差异明显,而原花青素和乌索酸其保护作用较为明显,原花青素在治疗酒精性肝损伤的效果要优于乌索酸的保护效果,具有较好的临床应用价值。 Objective To investigate the protective effect of two bioactive substances including procyanidin and ursolic acid on ethanol induced liver injury in mice.Methods The liver injury models were established in the same batch of mice.The mice were randomly divided into four groups.The mice in the two protection groups were given procyanidins and ursolic acid by gavage.After 2-week intragastric administration,the mice were were sacrificed.The livers were collected and homogenized and sliced.The levels of SOD,MDA,AST and ALT in serum and liver homogenate were measured.The injury of livers in different groups were observed and compared among groups.Results The comparison of the body weight of mice in four groups during the gavage period showed that there were significant difference in the changes of body weight of mice at differenct time points,and there were significant difference in the changes of body weight at 7d and 14d among groups(P<0.05).Moreover there was significant difference in body weight of mice at different days between the procyanidins group and ursolic acid group(P<0.05).There were significant differences in various indexes among the four groups[AST(F=9.836,P<0.01),ALT(F=7.539,P<0.01),MDA(F=28.829,P<0.01),SOD(F=8.491,P<0.01)].There were significant differences in the various indexes between the procyanidins group and ursolic acid group(P<0.05).In addition the liver injury in ursolic acid group was even more severe than that in procyanidins group.Conclusion There are significant differences in changes of various indexes in alcohol group.Moreover the protective effects of proanthocyanidins and ursolic acid are more obvious.The protective effects of procyanidins on alcoholic liver injury are superior to those by ursolic acid,which has preferable clinical application value.
