抗氧化剂锌和金属硫蛋白基因多态性与社区2型糖尿病的关系 被引量:2

Relationship between antioxidant zinc,metallothionein gene polymorphism and community type 2 diabetes mellitus

摘要 目的探析抗氧化剂锌(Zn)和金属硫蛋白(MT)基因多态性与社区2型糖尿病的关系。方法选取500例社区2型糖尿病患者纳入观察组,500例排除糖尿病的个体纳入对照组。所有受试者均于早晨空腹状态下检测血糖、血脂、锌离子、肝肾功能指标及rs2466295、rs11076160、rs11640851基因位点的基因型分布与频率。结果观察组血清TG、GLU高于对照组(P <0. 001),HDL低于对照组(P <0. 001)。观察组锌离子水平低于对照组,二者呈明显负相关(r=-0. 412,P=0. 020)。在rs11076161基因位点上,2组AA、AG、GG基因型分布的差异有统计学意义(P <0. 001);观察组A等位基因频率高于对照组(P <0. 001); rs11076161基因位点的基因多态性与糖尿病明显相关(OR=2. 778,P <0. 001)。在rs11640851基因位点上,2组AA、AG、GG基因型分差异有统计学意义(P <0. 001);观察组A等位基因频率(39. 00%)低于对照组(52. 00%)(P <0. 001); rs11640851基因位点的基因多态性与糖尿病明显相关(Or=0. 585,P <0. 001)。结论糖尿病与Zn T8及MT基因的多个多态性位点均有明显关联性,在rs2466295与rs11076161基因位点上,携带A等位基因的患病风险更大,在rs11640851基因位点上携带G基因的患病风险更大。 Objective To investigate the relationship between antioxidant zinc as well as metallothionein gene polymorphism and community type 2 diabetes mellitus.Methods A total of 500 patients with community type 2 diabetes were enrolled as observation group,and the other 500 patients who were excluded from diabetes were enrolled as control group.The blood glucose,blood lipids,zinc ions,liver and kidney function indexes,and the genotype distribution and frequency of rs2466295,rs11076160,rs11640851 gene loci were detected and compared between two groups.Results The serum levels of TG and GLU in observation group were significantly higher than those in control group(P<0.01),and the levels of HDL were significantly lower than those in control group(P<0.01).The levels of zinc in observation group were significantly lower than those in control group,there was a negative correlation between the two groups(r=0.412,P<0.05).In the rs11076161 gene locus,there was significant difference in the distribution of AA,AG and GG genotypes between two groups(P<0.01).The frequency of allele A in observation group was 62.80%,which was significantly higher than that(30.80%)in control group(P<0.005).There was a significant correlation between rs11076161 gene polymorphism and diabetes mellitus(OR=2.778,P<0.01).In the rs11640851 gene locus,there was significant difference in the distribution of AA,AG and GG genotypes between two groups(P<0.01).The frequency of allele A in observation group was 39.00%,which was significantly lower than that(52.00%)in control group(P<0.05).There was a significant correlation between rs11640851 gene polymorphism and diabetes mellitus(OR=0.585,P<0.01).Conclusion The diabetes and ZnT8,MT gene polymorphism sites are significantly correlated.In the rs2466295 and rs11076161 gene locus,the risk of suffering from the disease in patients carrying the A allele is greater,which in patients carrying the G gene at the rs11640851 gene locus is greater.
