WISP-1对单侧输尿管梗阻小鼠肾脏纤维化的作用研究 被引量:4

摘要 目的探讨Wnt1诱导信号通路蛋白1(WISP-1)在单侧输尿管梗阻(UUO)小鼠肾脏纤维化中的作用。方法通过腹腔注射WISP-1抗体作为处理组,以Ig G作为对照处理组; UUO和正常小鼠作为模型组及正常组。采用苏木素-伊红(HE)染色,高碘酸-雪弗(PAS)染色和Masson三色染色评估肾脏病理改变;免疫组织化学染色法(IHC)和蛋白质免疫印迹法(WB)检测小鼠肾脏WISP-1和胶原蛋白I(Collagen I)的表达。结果模型组及对照处理组较正常组,其梗阻侧肾组织出现不同程度的肾小球硬化、肾间质纤维化、细胞外基质及胶原纤维的沉积,而处理组上述病理损伤显著减轻。IHC和WB显示处理组WISP-1及Collagen I的表达较模型组及对照处理组明显下降。结论抗WISP-1抗体治疗能明显减轻UUO模型小鼠的肾脏纤维化。 To investigate the role of WNT1-inducible signaling pathway protein-1(WISP-1)in renal fibrosis of mice with unilateral ureteral obstruction(UUO).Anti-WISP-1 antibody was intraperitoneally injected into the UUO mice as a treatment group,while IgG-injected UUO mice were used as a control treatment group.The UUO and normal mice were used as a model group and a normal group.HE,PAS and Masson trichrome staining were used to evaluate the pathological changes of obstructed kidneys in the UUO mice.WB and IHC were used to detect the expression of WISP-1 and fibrosis marker of collagen I.Compared with the normal mice,different degrees of glomerulosclerosis,renal interstitial fibrosis,deposition of extracellular matrix and collagen fibers were observed in the UUO group and the control treatment group.The pathological damage of the kidneys in the anti-WISP-1 treated mice was significantly reduced.WB and IHC showed that anti-WISP-1 antibody treatment successfully inhibited the expression of WISP-1 and collagen I in the kidney of the treatment group compared to the UUO and control treatment groups.Anti-WISP-1 antibody treatment can significantly reduce renal fibrosis in UUO mice.
