摘要
目的探讨幽门螺旋杆菌(Hp)感染患儿胃黏膜组织中微小RNA(miR)-146a表达变化及与Toll样受体4(TLR4)/细胞核因子-B(NF-κB)信号通路的相关性。方法选取2014年2月至2016年12月在该院就诊的患儿107例,分为Hp感染组(n=58)和非感染组(n=49),原位杂交试验检测患儿胃黏膜组织中miR-146a表达,实时荧光定量PCR检测患儿胃黏膜组织中miR-146a、TLR4mRNA和NF-κB mRNA表达,酶联免疫吸附试验(ELISA)检测患儿胃黏膜组织中白细胞介素(IL)-6、IL-8、IL-10和肿瘤坏死因子(TNF)-α水平。结果 Hp感染组患儿胃黏膜组织中miR-146a阳性表达率为72.4%(42/58),非感染组为22.4%(11/49),差异有统计学意义(χ2=26.526,P=0.000);Hp感染组患儿胃黏膜组织中miR-146a、TLR4mRNA和NF-κB mRNA相对表达量均高于非感染组,差异均有统计学意义(P<0.05);Hp感染组患儿胃黏膜组织中IL-6、IL-8、IL-10和TNF-α水平均高于非感染组,差异均有统计学意义(P<0.05);Pearson相关分析显示,Hp感染组患儿胃黏膜组织中miR-146a相对表达量与TLR4mRNA相对表达量、NF-κB mRNA相对表达量、IL-6、IL-8、IL-10和TNF-α水平均呈正相关(P<0.05)。结论 Hp感染患儿胃黏膜组织中miR-146a呈高表达,可能通过调控TLR4/NF-κB信号通路介导的炎性反应参与了Hp感染致病过程。
Objective To investigate the expression change of miR-146a expression in gastric mucosal tissue and its correlation with TLR4/NF-κB signaling pathway in children patients with Helicobacter pylori(Hp)infection.Methods One hundred and seven children patients in the Zhengzhou Children′s Hospital from February 2014 to December 2016 were selected and divided into the Hp infection group(n=58)and non-Hp infection group(n=49).The expression of miR-146a in gastric mucosal tissues was detected by using in situ hybridization assay.Real-time quantitative PCR was used to detect the expressions of miR-146a,TLR4 mRNA and NF-κB mRNA in gastric mucosal tissues.The levels of IL-6,IL-8,IL-10 and TNF-αin gastric mucosal tissues were detected by ELISA.Results The positive expression rate of miR-146a in gastric mucosal tissues in the Hp infection group was 72.4%(42/58),while which in the non-Hp infection group was 22.4%(11/49),and the difference was statistically significant(χ2=26.526,P=0.000).The relative expression levels of miR-146a,TLR4 mRNA and NF-κB mRNA in gastric mucosal tissues in the Hp infection group were higher than those in the non-Hp infection group,and the differences were statistically significant(P<0.05).The levels of IL-6,IL-8,IL-10 and TNF-αin the Hp infection group were higher than those in the non-Hp infection group,and the differences were statistically significant(P<0.05).The Pearson correlation analysis showed that the relative expression level of miR-146a in gastric mucosal tissues in the Hp infection group was positively correlated with the relative expression level of TLR4 mRNA,the relative expression level of NF-κB mRNA and the levels of IL-6,IL-8,IL-10 and TNF-α(P<0.05).Conclusion miR-146a shows high expression in gastric mucosal tissues of children patients with Hp infection,which might be involved in the Hp pathogenic process by regulating the inflammatory response mediated by TLR4/NF-κB signaling pathway.
