摘要
利用聚T单链DNA为模板合成的荧光铜纳米粒子作为荧光探针,设计了一种简单、快速、灵敏的荧光生物传感方法用于乙酰胆碱酯酶活性的检测。验证了实验可行,对反应时间、硫代乙酰胆碱的浓度进行了优化,在最优条件下对乙酰胆碱酯酶的灵敏度进行了检测,检测限为0.036U·L-1,并证明了本实验具有良好的选择性。最后,探索了抑制剂浓度与体系荧光强度的关系,并且对比了他克林和加兰他敏抑制剂的抑制效率。
A simple,rapid and sensitive fluorescence biosensor was developed for the detection of acetylcholinesterase activity using fluorescent copper nanoparticles synthesized by poly-T single-stranded DNA as a fluorescent probe.It was proved that the experiment was feasible,the reaction time and the concentration of thioacetyl chloride were optimized.The sensitivity of acetylcholinesterase was tested under the optimal conditions.The detection limit was 0.036 U·L-1 and it was proved that the experiment had good selectivity.Finally,the inhibitory efficiency of tacrine and galanthamine was compared.
