结直肠癌中MACC1的表达及与淋巴结转移的相关性 被引量:8

Expression of MACC1 in colorectal carcinoma and its correlation with lymphatic metastasis

摘要 目的研究结肠癌转移相关基因-1(MACC1)在结直肠癌(CRC)中的表达及与淋巴结转移的相关性。方法免疫组化检测MACC1在77例CRC及其癌旁正常黏膜组织中的表达,免疫细胞化学检测不同转移潜能CRC细胞株的MACC1表达。结果 MACC1在CRC的阳性率为76.62%,显著高于癌旁黏膜的11.69%(χ2=65.83,P=0.00)。对无淋巴结和有淋巴结转移行单因素分析,分化程度、肿瘤大小、浸润深度、MACC1表达、神经侵犯和临床分期与淋巴结转移有关(P <0.05)。Logistic回归分析示MACC1表达、神经侵犯、浸润深度和临床分期是淋巴结转移的独立危险因素(P <0.05)。免疫细胞化学示MACC1在高淋巴结转移潜能的SW620中高表达,在低淋巴结转移潜能的SW480中低表达。结论 MACC1的异常高表达参与了CRC的恶性进展,是淋巴结转移的独立危险因素,具有重要临床价值。 Objective To investigate the expression of metastasis-associated in colon cancer-1(MACC1)in colorectal carcinoma(CRC)and its correlation with lymphatic metastasis.Methods MACC1 protein expression was detected in 77 cases of CRC tissues compared with the adjacent normal mucosa by immunohistochemistry assay,and the relationship between MACC1 and lymph node metastasis was analyzed.MACC1 expression was detected by immunocytochemistry assay in two CRC cell lines with different lymphatic potentials.Results The positive rate of MACC1 in CRC tissues was 76.62%,which was significantly higher than 11.69%in the adjacent normal mucosa(χ2=65.83,P=0.00).The single factor comparative analysis between none-metastasis group and metastasis group showed that the histological differentiation,tumor size,invasion depth,MACCl expression,nerve invasion and clinical stages were associated with lymph node metastasis(P<0.05,respectively).Logistic regression analysis showed that MACCl expression,nerve invasion,invasion depth and clinical stages were independent risk factors for lymph node metastasis(P<0.05,respectively).MACC1 protein expression was high in SW620 cell with high poten-tial for lymphatic metastasis,but was low in SW480 cell with low lymphatic metastasis.Conclusion Deregulation of MACCl may synergistically participate in the malignant progression of CRC.MACCl expression is an independent risk factor for cervical carcinoma pelvic lymph node metastasis,with an important clinical value.
