摘要
采用头孢曲松钠灌胃诱导小鼠肠道菌群失调,研究抗生素对小鼠肠道菌群组成及代谢的影响。通过宏基因组分析小鼠肠道菌群组成;气相色谱法测定小鼠粪便中短链脂肪酸的质量分数;ELISA试剂盒测定小鼠血清中脂多糖浓度。抗生素灌胃后可引起小鼠粪便中厚壁菌门增加,拟杆菌门减少;乙酸、丙酸和丁酸等短链脂肪酸质量分数下降;血清中脂多糖浓度升高。抗生素造成的小鼠肠道菌群紊乱,可导致有害菌的丰度增加并抑制益生菌生长;肠道菌群紊乱可导致短链脂肪酸质量分数下降及脂多糖浓度升高,促进炎症的发生。
In the present paper,the effects of antibiotic on intestinal microbiota communities and metabolism of mice were investigated by using an animal model of intestinal flora disorder induced by ceftriaxone sodium in mice.The influence of ceftriaxone sodium on the intestinal microbiota communities of mice was examined by high-through sequencing.Short chain fatty acids(SCFAs)in mice feces were detected by gas chromatography(GC).Concentrations of lipopolysaccharide(LPS)were detected by LPS ELISA kit.The most changes in intestinal microbiota communities of mice induced by ceftriaxone sodium were Firmicutes and Bacteroidetes.Ceftriaxone sodium caused a decrease of the contents of SCFAs,but an increase of LPS.These data showed that the antibiotic administered to mice alter the bacterial communities by promoting the production of harmful bacteria and inhibiting the growth of beneficial bacteria;disorder of intestinal flora can reduce SCFAs concentrations and accelerate the production of LPS,and promote the inflammation.
