摘要
本研究采用HPLC-DAD法,在单因素试验的基础上,用响应曲面法(RSM)对影响稻谷中叶黄素提取效果的四氢呋喃用量、KOH甲醇溶液质量浓度和提取温度三个因素进行优化,建立各因素与叶黄素提取量关系的数学模型。响应曲面法优化稻谷中叶黄素最佳提取方法为四氢呋喃15.5mL、KOH甲醇溶液质量浓度0.1g/mL、提取温度51℃,稻谷中叶黄素提取量为(1.63±0.03)μg/g。运用HPLC-DAD及响应曲面法可有效缩短稻谷中叶黄素提取时间,为高效提取稻谷中的叶黄素提供了参考。
On the basis of single factor experiment,response surface methodology(RSM)was used to optimize the three factors affecting the extraction of lutein from rice,namely the amount of tetrahydrofuran,the concentration of KOH methanol solution and the extraction temperature.The mathematical model of the relationship between each factor and the extraction amount of lutein was established.Response surface methodology was used to optimize the extraction of lutein from paddy.The optimum extraction methods were as follows:the amount of tetrahydrofuran was 15.5mL,the concentration of KOH methanol solution was 0.10 g/mL,the extraction temperature was 51℃,and the extraction amount of lutein from paddy was(1.63±0.03)ug/g.The extraction time of lutein in rice can be effectively shortened by using HPLC-DAD and response surface methodology,which provides a reference for efficient extraction of lutein in rice.
