云南甘蔗病毒性病害的分子鉴定

Molecular identification of virus diseases in sugarcane in Yunnan

【目的】了解5种常见甘蔗病毒性病害在云南的发生情况,明确其病原病毒的种类和分布,为制定云南地区甘蔗病毒性病害的防治策略提供理论依据。【方法】利用PCR和RT-PCR对采集自云南保山、临沧和德宏3个甘蔗种植区的113份样品进行甘蔗花叶病毒(Sugarcane mosaic virus,SCMV)、高粱花叶病毒(Sorghum mosaic virus,SrMV)、甘蔗线条花叶病毒(Sugarcane streak mosaic virus,SCSMV)、甘蔗黄叶病毒(Sugarcane yellow leaf virus,SCYLV)和甘蔗杆状病毒(Sugarcane bacilliform virus,SCBV)的分子鉴定。【结果】113份甘蔗样品中有89份检出病毒,病毒检出率为78.8%,检出的病毒种类有SrMV、SCSMV、SCYLV和SCBV,检出率分别为5.3%、11.5%、20.4%和70.8%,未检出SCMV。30份样品存在病毒复合侵染现象,复合侵染率为26.5%,其中有27份样品为2种病毒复合侵染,占23.9%,3份样品发生3种病毒复合侵染,占2.7%;未发现4种或5种病毒复合侵染的现象。【结论】目前云南地区甘蔗病毒性病害由SrMV、SCSMV、SCYLV和SCBV引起,其中SCBV广泛分布,SrMV、SCSMV和SCYLV具有一定程度的分布;病毒复合侵染现象明显,以2种病毒复合侵染为主。

【Objective】The occurrence of five common sugarcane virus diseases and the species and distribution of their pathogenic viruses in Yunnan were studied to provide theoretical basis for the development of control strategies for sugarcane virus diseases in Yunnan.【Method】A total of 113 sugarcane samples collected from three sugarcane growing areas(Baoshan,Lincang and Dehong)in Yunnan were detected the molecular identification of five viruses,namely,Sugarcane mosaic virus(SCMV),Sorghum mosaic virus(SrMV),Sugarcane streak mosaic virus(SCSMV),Sugarcane yellow leaf virus(SCYLV)and Sugarcane bacilliform virus(SCBV),via PCR and RT-PCR methods.【Result】A total of 89 out of the 113 sugarcane samples were detected viruses positive with a detection rate of 78.8%.The detected virus species were SrMV,SCSMV,SCYLV and SCBV,and the detection rates were 5.3%,11.5%,20.4%and 70.8%respectively.SCMV was not detected in all samples.Thirty samples were co-infected with different viruses,and the co-infection rate was 26.5%.Among them,twenty-seven samples were co-infected with two viruses,accounting for 23.9%,and three samples were co-infected with three viruses,accounting for 2.7%.No samples were co-infected with four or five viruses.【Conclusion】The sugarcane virus diseases are caused by SrMV,SCSMV,SCYLV and SCBV in Yunnan.SCBV widely distributes in Yunnan,and SrMV,SCSMV and SCYLV also distribute in some parts of Yunnan.Co-infection with several viruses,especially two viruses,is common in sugarcane in Yunnan.
