茶树炭疽病病原菌的分离与鉴定 被引量:11

摘要 为茶炭疽病的快速诊断及防治提供理论依据,采用常规组织分离、伤口回接法对比感病症状,并通过形态学特征结合多基因系统发育分析方法,探究遵义银柜山茶区福鼎大白茶炭疽病病原菌的种类及致病力。结果表明:分离得到茶树炭疽病菌株1株(ygs03-1),该菌株在PDA平板上25℃黑暗条件下培养,菌落正面呈白色或灰白色海绵状隆起,背面黑褐色;气生菌丝发达,透明有隔;分生孢子梗透明、有隔膜;产孢细胞透明,圆柱形或椭圆形;分生孢子长椭圆或椭圆形,少部分为花生形;分生孢子附着孢圆形或不规则形,边缘呈黑色。ygs03-1可通过伤口侵染茶树叶片,且病症与采集病样病症相似。经系统进化树聚类分析,分离菌株ygs03-1和Colletotrichum camelliae聚为一类,相似度达97%。结合该菌株的形态特征与分子鉴定结果,确定菌株ygs03-1为刺盘孢炭疽菌(Colletotrichum camelliae)。 The species and pathogenicity of Colletotrichum camelliae pathogen to Fudingdabaicha in Yinguishan TEA District,Zunyi were investigated by using usual tissue isolation,wound healing method,morphological feature and polygenic system analysis to provide the theoretic basis for rapid diagnosis and control of C.camelliae.Result:One C.camelliae strain(ygs03-1)is isolated.The color of the colony on front and reverse side is white or grey white and black brown respectively when ygs03-1 strain is cultured on PDA plate at 25℃under dark condition.The flourishing aerial hyphae are transparent with diaphragms.The transparent conidiophore has diaphragms.The shape of the transparent generator cells is cylinder or oval.The shape of most conidia is prolate ellipse or ellipse and the shape of a few conidia is peanut-shaped.The shape of conidia′appressorium is roundness or irregular and the edge color is black.The ygs03-1 strain can infect leaves of tea tree by the wound and its symptoms is similar to the collected disease sample.The phylogenetic tree clustering analysis shows that the similarity between ygs03-1 strain and C.camelliae reaches 97%.The ygs03-1 strain is identified as C.camelliae by its morphological feature and molecular identification.
