摘要
为杨梅炭疽病生物农药的开发提供新的生防菌资源,采用解磷选择性培养基分离及平板对峙筛选相结合的方法从杨梅种植土壤中筛选杨梅炭疽病菌拮抗菌株,通过菌落直径法验证其发酵液的抑菌特性,并利用形态学与分子生物学相结合的方法对拮抗菌株进行鉴定。结果表明:从杨梅根际土壤中分离获得52株解磷真菌,其中菌株YMMF150101对杨梅炭疽病菌有拮抗作用,平板对峙抑制率达81.6%;其发酵液及无菌发酵滤液均对靶标菌生长有抑制作用,抑制率分别达84.73%和52.53%;根据形态学特征结合其分子系统发育学鉴定,YMMF150101为米曲霉菌(Aspergillus oryzae)。
An antagonistic strain against anthracnose was isolated from Morella rubra planting soil by culture of phosphate-solubilizing selective medium and plate confrontation,the inhibition characteristics of the fermentation broth of the antagonistic strain was tested by the colony diameter method and the isolated antagonistic strain was identified by the methods of morphology and molecular biology to provide the new bio-control fungi for development of biopesticides against anthracnose in M.rubra.Result:52 phosphate-solubilizing fungi strains are isolated from rhizosphere soil of M.rubra and the plate confrontation inhibition rate of YMMF150101 strain with an antagonism against M.rubra anthracnose reaches 81.6%.The inhibition rate of its fermentation broth and sterile fermentation filtrate against growth of target fungi is 84.73%and 52.53%respectively.YMMF150101 strain is identified as Aspergillus oryzae by its morphological characteristics and molecular phylogeny.
