摘要
目的比较拉布拉多犬牙槽骨干细胞(Al-BMSC)与髂骨骨髓间充质干细胞(I-BMSC)促进犬种植体近中临界骨缺损修复以及与种植体实现骨结合的能力。方法构建拉布拉多犬下颌骨种植体近中临界骨缺损动物模型,分别植入β-TCP+Al-BMSC、β-TCP+I-BMSC及单纯β-TCP。术后4周、8周、10周分别注射荧光标记物四环素、钙黄绿素与茜素红,术后12周获取标本进行荧光标记组织学分析以及硬组织切片组织学分析。结果荧光标记组织学分析结果显示,β-TCP+Al-BMSC组和β-TCP+I-BMSC组的荧光表达量均比单纯β-TCP组多(P <0. 05)。β-TCP+Al-BMSC组与β-TCP+I-BMSC组的荧光表达量差异无统计学意义。硬组织切片组织学分析结果显示,β-TCP+Al-BMSC组和β-TCP+I-BMSC组的新骨形成面积均比单纯β-TCP组多(P<0. 05),β-TCP+Al-BMSC组和β-TCP+I-BMSC组间的新骨形成面积差异无统计学意义;β-TCP+I-BMSC组种植体周围骨结合率高于β-TCP+Al-BMSC组,差异具有统计学意义(P <0. 05)。结论 Al-BMSC和I-BMSC均可促进种植体近中临界骨缺损的修复,两者的成骨能力没有明显差异。IBMSC可以促进更多的种植体周围骨结合。
Objective To compare the osteogenic potential of iliac bone marrow mesenchymal stem cells(I-BMSCs)with alveolar bone marrow MSCs(Al-BMSCs)when combined withβ-TCP in critical-size bone defect around implants as well as to investigate the osseointegration between tissue-engineered bone and dental implants in dogs.Methods To construct canine model of mandible critical-size bone defect.Cell-seeded scaffolds were put in defects,which were created in mandibular premolars area with simultaneous implant placement.The experiment was divided into 3 groups:β-TCP+Al-BMSCs group,β-TCP+I-BMSCs group,β-TCP group.Sequence fluorescence labeling was operated at 4 weeks,8 weeks and 10 weeks after the surgery.12 weeks after the surgery,the specimen was harvested and analyzed histologically.Results More new bone was detected in theβ-TCP+Al-BMSCs group andβ-TCP+I-BMSCs group than that in theβ-TCP group(P<0.05).There was no statistical difference of the new bone formation between theβ-TCP+Al-BMSCs group andβ-TCP+I-BMSCs group,while BIC with obvious statistical difference was higher in theβ-TCP+I-BMSCs group than that in theβ-TCP+Al-BMSCs group(P<0.05).Conclusion Both Al-BMSCs and I-BMSCs have the ability of osteogenesis.More BIC could be obtained by using I-BMSCs.
