利拉鲁肽对糖尿病小鼠血糖和血脂影响的研究 被引量:3

摘要 目的探讨利拉鲁肽对糖尿病小鼠血糖和血脂等指标的影响作用。方法将40只雄性小鼠随机分为3组:对照组(n=10),模型组(B组,n=15),模型+利拉鲁肽组(A组,n=15)。对照组腹腔注射等体积0. 1 mol/L柠檬酸缓冲液,持续5 d。所有模型组小鼠给予腹腔注射70 mg/kg现配的链脲佐菌素,持续5 d。注射结束后1周后检测随机血糖水平> 16. 7 mmol/L视为建模成功。A、B两组分别有2只小鼠未建模成功。随后A组小鼠腹腔注射200 g/kg利拉鲁肽,持续4周。检测糖耐量、血脂、肝功能等指标。结果与对照组相比,B组小鼠对血糖的调节能力降低,糖耐量受损。而利拉鲁肽可改善这种糖耐量受损。A、B两组小鼠的甘油三酯(TG)、总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)水平显著高于对照组小鼠,其中A组显著低于B组(P <0. 05),A、B两组小鼠的高密度脂蛋白胆固醇(HDL-C)水平显著低于对照组小鼠,其中A组显著高于B组(P <0. 05); A、B两组小鼠的天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、总胆汁酸(TBA)和血尿酸(UA)水平显著高于对照组小鼠,且研究A组显著低于B组(P <0. 05)。结论利拉鲁肽能够改善糖尿病小鼠的糖耐量、血脂和肝功能各项指标的水平,治疗效果显著,可作为糖尿病治疗的潜在药物推广使用。 Objective To investigate the effects of liraglutide on blood glucose and blood lipids in diabetic mice.Methods 40 male mice were randomly divided into 3 groups,control group(n=10),model group(B group=15)and model group combined with liraglutide(A group=15).The control group was intraperitoneally injected with an equal volume of 0.1 mol/L citrate buffer for 5 days.All mice in the model group were given intraperitoneal injection of streptozotocin at 70 mg/kg for 5 days.Detection of random blood glucose level>16.7 mmol/L at 1 week after injection was deemed as successful modeling.2 mice in A and B groups respectively were not successfully modeled.Subsequently,mice in A group were intraperitoneally injected with 200 g/kg liraglutide for 4 weeks.The indicators of glucose tolerance,blood lipid and liver function were tested.Results Compared with the control group,the mice in B group had a lower ability to regulate blood sugar and impaired glucose tolerance.Liraglutide can improve this impaired glucose tolerance.The level of TC,TG and LDL-C in A and B groups were significantly higher than those in control group,and the A group was significantly lower than in B group(P<0.05),the level of HDL-C in A and B groups were significantly lower than those in control group,and the A group was significantly higher than that in B group(P<0.05).The ALT,AST,TBA and UA levels in A and B groups were significantly higher than those in control group,and the A group was significantly lower than in B group(P<0.05).Conclusion Liraglutide can improve glucose tolerance and the levels of blood lipid and liver function in diabetic mice,and has significant therapeutic effect.Liraglutide can be widely used as a potential drug for the treatment of diabetes.
