叶绿醇对小鼠骨骼肌代谢酶活性和肌纤维类型的影响 被引量:6

摘要 本试验旨在探讨饲粮中添加叶绿醇对小鼠骨骼肌代谢酶活性和肌纤维类型的影响。试验选择40只3周龄雄性昆明小鼠,随机分为4组,每组10只,单笼饲养。试验期分别在小鼠饲粮中添加0(对照组)、0.05%和0.50%的叶绿醇,并以添加0.48%硬脂酸为阳性对照组。试验测定小鼠平均体重,腓肠肌相对重量,腓肠肌中琥珀酸脱氢酶(SDH)、己糖激酶(HK)和乳酸脱氢酶(LDH)活性及甘油三酯(TG)含量,并通过ATP酶染色法研究肌纤维类型。试验期28 d。结果表明:1)与对照组相比,饲粮中添加0.50%叶绿醇,小鼠试验前2周平均体重显著降低(P<0.05),腓肠肌相对重量显著增加(P <0.05)。2)与对照组相比,饲粮中添加0.50%叶绿醇显著降低小鼠腓肠肌中TG含量(P <0.05),显著提高腓肠肌中HK活性(P <0.05);饲粮中添加0.05%叶绿醇显著提高小鼠腓肠肌中SDH活性(P<0.05)。3)与对照组相比,饲粮中添加0.50%叶绿醇显著提高小鼠腓肠肌中Ⅰ型肌纤维比例(P<0.05)。综合以上结果表明,饲粮中添加叶绿醇可以提高小鼠腓肠肌相对重量,提高腓肠肌中SDH和HK的活性,增加腓肠肌中Ⅰ型肌纤维比例。 This study was aimed to investigate the effects of phytol on metabolic enzyme activity of skeletal muscle and muscle fiber type of mice.Forty 3-week-old Kunming male mice were randomly divided into 4 groups with 10 in each group,raised in separate cages,and fed diets containing 0(control group),0.05%,0.50%phytol and 0.48%stearic acid(positive control group).The average body weight,relative weight of the gastrocnemius,activities of succinodehydrogenase(SDH),hexokinase(HK),and lactic dehydrogenase(LDH)and triglyceride(TG)content of mice were detected,and ATPase staining was conducted to observe muscle fiber type of mice.The experiment lasted 28 d.The results showed as follows:1)compared with the control group,adding 0.50%phytol significantly decreased the average body weight of mice in the first 2 weeks(P<0.05),and significantly increased the relative weight of the gastrocnemius(P<0.05).2)Compared with the control group,adding 0.50%phytol significantly reduced TG content in the gastrocnemius of mice(P<0.05),and significantly increased HK activity in the gastrocnemius(P<0.05);adding 0.05%phytol significantly increased SDH activity in the gastrocnemius of mice(P<0.05).3)Compared with the control group,0.50%phytol significantly increased the proportion ofⅠmuscle fiber type in the gastrocnemius of mice(P<0.05).Above all,these results indicate that phytol could increase the relative weight of the gastrocnemius,the activities of SDH and HK,and the proportion ofⅠmuscle fiber type in the gastrocnemius of mice.
