摘要 目的:酶联免疫吸附测定法(ELISA)和薄层层析法(TLC)是检测食品中黄曲霉素B_1的常规方法。通过比较ELISA和TLC检测茶叶黄曲霉素B_1的准确性,确定作为评价茶叶质量及安全性的手段。方法:以市售的普洱茶和毛尖茶为样品,添加黄曲霉毒素B_1标样制成待检样品。分别利用方法学原理对ELISA和TLC检测茶叶样品中黄曲霉毒素B_1进行科学验证。结果:酶联免疫法相对于薄层层析法,在精密度、重现性、稳定性、特异性等方面存在着特异性不好,误差较大等不足。TLC对普洱茶及毛尖茶中未检出黄曲霉素。结论:由于茶叶中成分比较复杂,ELISA不适用于茶叶黄曲霉毒素B_1检测,TLC可用于检测茶叶黄曲霉素B_1,但也存在操作复杂、费时等不足。目前,TLC用于茶叶安全性监测以及对进一步开发更准确、快捷、适用的黄曲霉毒素B_1检测方法,辅助评价意义重大。 Objecktive:Enzyme-linked immunosorbent assay(ELISA)and thin layer chromatography(TLC)are conventional methods for detecting aflatoxin B1in foods.The accuracy of aflatoxin B1in tea was determined by comparing ELISA and TLC,and it was determined as a means to evaluate the quality and safety of tea.Methods:The commercially available Pu-erh tea and Maojian tea were used as samples,and the aflatoxin B1standard was added to prepare a sample to be tested.The methodological principle was used to verify the aflatoxin B1in tea samples by ELISA and TLC.Results:Compared with TLC,ELISA has some shortcomings,such as poor specificity and large error in precision,reproducibility,stability and specificity.Aflatoxin was not detected in Pu’er tea and Maojian tea by TLC method.Conclusion:Due to the complexity of the components in tea,ELISA is not suitable for the detection of aflatoxin B1in tea.TLC can be used to detect aflatoxin B1in tea,but it also has the disadvantages of complicated operation and time-consuming.At present,it is of great significance for tea safety monitoring and for the further develop a more accurate,fast and applicable method for aflatoxin B1detection.
