摘要
目的比较4种不同浓度水凝胶在小鼠脑、肝、肾组织透明化中的应用效果。方法根据丙烯酰胺(A)、甲叉双丙烯酰胺(B)、多聚甲醛(P)的终浓度比,制备A4B4P4(A∶B∶P=4%∶0. 05%∶4%)、A2B2P4(A∶B∶P=2%∶0. 025%∶4%)、A1B1P4(A∶B∶P=1%∶0. 0125%∶4%)、A4B4P0(A∶B∶P=4%∶0. 05%∶0)水凝胶。选取8只昆明小鼠随机分为4组,每组2只,分别采用A4B4P4、A4B4P0、A2B2P4、A1B1P4水凝胶进行灌注,然后行组织固定及透明化处理。计算经4种水凝胶处理后小鼠脑、肝、肾组织的相对透明度及蛋白损失量。结果 A2B2P4组、A1B1P4组小鼠脑组织相对透明度均高于A4B4P4、A4B4P0组(均P <0. 05); A1B1P4组小鼠肝组织的相对透明度高于其他3组(均P <0. 05); A2B2P4、A1B1P4组小鼠肾组织的相对透明度均高于A4B4P4、A4B4P0组(均P <0. 05)。A4B4P0组小鼠脑、肝、肾组织蛋白损失量均大于其他3组(均P <0. 05),A2B2P4组小鼠肝、肾组织蛋白损失量均低于A1B1P4组(均P <0. 05)。结论 A2B2P4、A1B1P4水凝胶在小鼠脑组织透明化中的应用效果最佳,A1B1P4、A2B2P4水凝胶分别在小鼠肝组织、肾组织透明化中的应用效果最佳。
Objective To compare the application efficacy among four hydrogels with different concentrations in mouse brain,liver and kidney tissues clearing.Methods According to the final concentration ratio of acrylamide(A),bisacrylamide(B)and paraformaldehyde(P),hydrogels A4B4P4(A∶B∶P=4%∶0.05%∶4%),A2B2P4(A∶B∶P=2%∶0.025%∶4%),A1B1P4(A∶B∶P=1%∶0.0125%∶4%)and A4B4P0(A∶B∶P=4%∶0.05%∶0)were prepared.Eight Kunming mice were selected and divided into4groups,with2mice in each group.The mice in the four groups were perfused with hydrogels A4B4P4,A4B4P0,A2B2P4and A1B1P4separately,then tissue fixation and tissue clearing were conducted.The relative transparency and protein loss of mouse brain,liver and kidney tissues were calculated after treatment with four hydrogels.Results The relative transparency of mouse brain tissues in the A2B2P4group and A1B1P4group was higher than that in the A4B4P4group and A4B4P0group(all P<0.05);the relative transparency of mouse liver tissues was higher in the A1B1P4group than that in the other three groups(all P<0.05);the relative transparency of mouse kidney tissues in the A2B2P4group and A1B1P4group was higher than that in the A4B4P4group and A4B4P0group(all P<0.05).The protein loss of mouse brain,liver and kidney tissues in the A4B4P0group was greater than that in the other three groups(all P<0.05),the protein loss of mouse liver and kidney tissues in the A2B2P4group was less than that in the A1B1P4group(all P<0.05).Conclusion The application of A2B2P4and A1B1P4hydrogels obtains optimal efficacy in mouse brain tissues clearing.The application of A1B1P4and A2B2P4hydrogels obtains optimal efficacy in mouse liver and kidney tissues respectively.
