解毒化瘀方对S180荷瘤小鼠5-氟尿嘧啶化疗的增效减毒作用研究

Study on the synergistic effect of Jiedu Huayu Decoction on 5-fluorouracil chemotherapy in S180 tumor-bearing mice

目的探讨解毒化瘀方对S180荷瘤小鼠5-氟尿嘧啶(5-Fu)化疗效果及毒副作用的影响。方法制备S180荷瘤小鼠的实体瘤动物模型,将造模成功小鼠随机分为生理盐水组、10 mg/kg 5-Fu组、20 mg/kg 5-Fu组、1 200 mg/kg中药组、2 400 mg/kg中药组、10 mg/kg 5-Fu联合1 200 mg/kg中药组、10 mg/kg 5-Fu联合2 400 mg/kg中药组,每组14只。接种24 h后开始干预13 d,5-Fu腹腔注射,隔日1次;中药灌胃,每日1次;生理盐水组灌胃中药同等体积的生理盐水。观察各组小鼠的体质量变化,末次给药后24h处死各组小鼠,称取肿瘤、胸腺及脾脏质量,计算肿瘤抑制率、胸腺指数及脾脏指数。结果干预13 d后,20 mg/kg 5-Fu组小鼠体质量明显低于其他组(P均<0. 05),其他各组小鼠体质量比较差异均无统计学意义(P均> 0. 05)。单纯5-Fu组、5-Fu联合中药组肿瘤质量均明显低于生理盐水组和单纯中药组(P均<0. 05),20 mg/kg 5-Fu组、5-Fu联合中药组肿瘤抑制率均明显高于10 mg/kg 5-Fu组(P均<0. 05),且10 mg/kg 5-Fu联合1 200 mg/kg中药组肿瘤抑制率更高(P均<0. 05)。单纯5-Fu组胸腺指数均明显低于生理盐水组(P均<0. 05),2 400 mg/kg中药组胸腺指数明显高于生理盐水组(P <0. 05),其他组胸腺指数与生理盐水组比较差异均无统计学意义(P均> 0. 05)。20 mg/kg 5-Fu组脾脏指数明显低于生理盐水组(P <0. 05),其他组脾脏指数比较差异均无统计学意义(P均> 0. 05)。结论解毒化瘀方单独使用对S180荷瘤小鼠肿瘤生长无抑制作用,与5-Fu联合使用具有明显的增效减毒作用。

Objective It is to investigate the effects of Jiedu Huayu Decoction on the curative effect and side effect of 5-fluorouracil(5-Fu)chemotherapy in S180 tumor-bearing mice.Methods The solid tumor model of S180 tumor-bearing mice was prepared.The successful mice were randomly divided into normal saline group,10 mg/kg 5-Fu group,20 mg/kg 5-Fu group,1 200 mg/kg Chinese medicine group and 2 400 mg/kg Chinese medicine group,10 mg/kg 5-Fu combined with 1 200 mg/kg Chinese medicine group,2 400 mg/kg Chinese medicine group,10 mg/kg 5-Fu plus 1 200 mg/kg Chinese medicine group,10 mg/kg 5-Fu plus 2 400 mg/kg Chinese medicine group,14 rats in each group.After 24 hours of inoculation,the intervention was started for 13 days,5-Fu was intraperitoneally injected once every other day;Chinese medicine was intragastrically administered once a day;the normal saline group was given the same volume of normal saline.The body weight changes of of mice in each group were observed.The mice in each group were sacrificed in 24 hours after the last administration.The tumor,thymus and spleen mass were weighed,and the tumor inhibition rate,thymus index and spleen index were calculated.Results After 13 days of intervention,the body weight of mice in the 20 mg/kg 5-Fu group was significantly lower than that in the other groups(P<0.05).There was no significant difference in body weight between the other groups(P>0.05).The tumor masses of the 5-Fu group and the 5-Fu plus Chinese medicine group were significantly lower than those in the normal saline group and the simple Chinese medicine group(P<0.05).The tumor inhibition rates of the 20 mg/kg 5-Fu group and the 5-Fu combined Chinese medicine group were higher than that in 10 mg/kg 5-Fu group(P<0.05),and the rate was even higher in 10 mg/kg 5-Fu combined with 1200mg/kg Chinese medicine group(P<0.05).The thymus index of the 5-Fu group was significantly lower than that of the saline group(P<0.05).The thymus index of the 2 400 mg/kg Chinese medicine group was significantly higher than that of the normal saline group(P<0.05).The difference in the thymus index of other groups were not different compared with normal saline group(P>0.05).The spleen index of 20 mg/kg 5-Fu group was significantly lower than that of normal saline group(P<0.05).There was no significant difference in spleen index among the other groups(P>0.05).Conclusion Jiedu Huayu Decoction used alone has no inhibitory effect on tumor growth of S180 tumor-bearing mice,and it has obvious effects for efficacy enhancing and toxicityreducing when combined with 5-Fu.
