摘要
目的探讨秋水仙碱长期干预高脂饮食大鼠的安全性。方法将30只SPF级健康雄性8周龄SD大鼠随机分为3组,各10只。对照组大鼠采用基础饲料喂养+生理盐水管饲喂养8周;高脂组大鼠采用按比例配方的高脂喂养+生理盐水管饲喂养8周,观察组大鼠采用按比例配方的高脂喂养并给予秋水仙碱[0.02 mg/(kg·d)]管饲喂养8周,比较三组大鼠的骨髓抑制情况和免疫功能。结果观察组的白细胞、红细胞、血红蛋白的骨髓抑制发生率高于对照组、高脂组,差异均有统计学意义(P<0.05)。观察组的CD3、CD8、CD56、CD4/CD8高于对照组、高脂组,差异有统计学意义(P<0.05)。观察组斑块内浸润的巨噬细胞个数为(17.93±3.87)%,低于高脂组的(22.98±2.12)%,差异有统计学意义(P<0.05),与对照组的(16.78±2.98)%比较,差异无统计学意义(P>0.05)。结论长期应用秋水仙碱干预高脂饮食大鼠会引起大鼠的骨髓抑制和免疫功能紊乱,该研究结果也提示临床治疗应综合考虑,进行全面评估,以提高治疗效果。
Objective To explore the safety of Colchicine for long-term intervention in rats fed with high-fat diet.Methods Thirty healthy and male rats(SPF grade)were randomly divided into the3groups,and there were10rats in each group.Rats in the control group were fed with basal feed and saline for8weeks.Rats in the high-fat group were given proportionally formulated high-fat feeding+saline tube feeding for8weeks.The rats in the observation group were given proportionally formulated high-fat feeding+Colchicine(0.02mg/[kg·d])tube feeding for8weeks.Bone marrow suppression and immune function were compared in rats among the three groups.Results The incidence of myelosuppression of leukocytes,erythrocytes,and hemoglobin in the observation group was higher than that of the control group and high-fat group reapectively,and the differences were statistically significant(P<0.05).The CD3,CD8,CD56,and CD4/CD8in the observation group was higher than that of the control group and high-fat group reapectively,and the differences were statistically significant(P<0.05).The number of infiltrating macrophages in plaque of the observation group was(17.93±3.87)%,which was significantly lower than that of the high-fat group([22.98±2.12]%),and the difference was statistically significant(P<0.05),and there was no significant difference between the observation group and control group([16.78±2.98]%)(P>0.05).Conclusion Long-term use of Colchicine in rats with high-fat diet can cause myelosuppression and immune dysfunction.The results also suggest that clinical treatment should be considered comprehensively and evaluated comprehensively in order to improve the therapeutic effect.
