摘要
目的探讨地奥司明预处理对新西兰白兔移植供心的保护作用及机制。方法选取健康成年新西兰大白兔36只,按简单随机抽样方法分为对照组、地奥司明预处理组、地奥司明+二乙基二硫代氨基甲酸钠(DDTC)预处理组。对照组正常饮食。地奥司明预处理组在实验开始前10 d予以地奥司明(80 mg/kg)灌胃,2次/d。地奥司明+DDCT预处理组在实验开始前10 d予以地奥司明(80 mg/kg)灌胃,2次/d,并在行同种异位心移植前15 min,于兔耳缘静注射DDTC(300 mg/kg)。比较对照组、地奥司明预处理组、地奥司明+DDTC预处理组中新西兰兔心脏移植后的心肌酶、凋亡指数、心肌组织含水量,并观察镜下心肌组织。结果地奥司明预处理组供心复跳后30、60、90 min心肌酶指标、心肌细胞凋亡指数和心肌组织含水量较另两组低(P均<0.05),镜下观察心肌组织结构较完好;地奥司明+DDTC预处理组各项指标较对照组均有降低,但差异无统计学意义(P均>0.05),且镜下心肌结构破坏严重。结论地奥司明预处理对家兔移植供心有保护作用,其机制与其提高了机体抗氧自由基能力有关。
Objective To investigate the protective effect and mechanism of dioximine pretreatment on transplanted donor heart in New Zealand white rabbits.Methods Thirty-six healthy adult New Zealand white rabbits are randomly divided into the control group,dioximine pretreatment group,and dioximine+diethyldithiocarbamate sodium(DDTC)pretreatment group.The rabbits in the control group were fed normally.The rabbits in the dioximine pretreatment group were given dioximine80mg/kg by stomach tube,twice a day,10days before the experiment.The rabbits in the dioximine+DDTC pretreatment group were given dioximine80mg/kg by stomach tube,twice a day,10days before the experiment,and DDTC300mg/kg was injected into the edges of ears at15min before heart transplantation.The myocardial enzyme,apoptosis index and water content in the myocardial tissues were compared among groups,and the myocardial tissues were observed under the microscope.Results Compared with the other two groups,the myocardial enzyme index,myocardial apoptosis index,and the water content of myocardial tissues decreased in the dioximing pretreatment group at30,60,and90min after the donor heart resuscitated(all P<0.05),and the myocardial tissue structure under microscope was better.All the indexes in the dioxomine+DDTC pretreatment group were lower than those in the control group,but the difference was not statistically significant(all P>0.05),and the myocardial structure under microscope was severely damaged.Conclusion Dioximine pretreatment has a protective effect on rabbit transplanted donor heart,and its mechanism is related to the improvement of antioxidant free radical ability.
