摘要
[目的]建立甘薯黑斑病菌快速大量产孢体系,为甘薯黑斑病品种抗性鉴定以及药剂生物测定奠定基础。[方法]采用3种方法制备甘薯黑斑病菌甘薯长喙壳菌(Ceratocystis fimbriata Ellis et Halsted)孢子悬浮液,研究甘薯黑斑病菌快速大量产孢方法。[结果]洗涤PDA和洗涤薯片培养5~6 d后镜检孢子悬浮液,一个视野内孢子数分别约为93、95个。利用PS培养液培养1、2、4 d后镜检孢子数分别为211、136、137个。[结论]采用PS液体培养基在特定培养条件下摇培1 d即可获得大量高浓度孢子悬浮液,极大地缩短了黑斑病菌产孢时间,显著提高了产孢量,且不易污染。
[Objective]To establish rapid sporulation system for Ceratocystis fimbriata,and provide an important foundation for resistant cultivar identification and pesticide bioassay of Ceratocystis fimbriata.[Method]Three methods were used for sporulation of C.fimbriata to study methods for rapid sporulation of C.fimbriata.[Result]5-6days after cultured,in one field of view there were93and95spores in the control group1and the control group2,respectively.However,the numbers of spores were211,136and137after1day,2days and4days cultured in PS medium,respectively.[Conclusion]A large amount of high concentration spores suspension can be obtained by shaking culture1day in PS liquid medium under a specific culture condition,which can greatly shorten the sporulation time of C.fimbriata,also it was not easy to be polluted.
