双歧杆菌对高糖高脂饮食与STZ诱导小鼠肠道菌群及肾功能影响的研究 被引量:5

摘要 目的探讨双歧杆菌干预对高糖饮食与STZ诱导小鼠肠道菌群的影响。方法 30只雌性C57BL/6J小鼠随机分为正常对照组、模型组和双歧杆菌干预组,每组10只,正常组给予标准饲料,模型组给予高糖饲料和STZ注射,双歧杆菌干预组给予高糖饲料、STZ注射同时给予双歧杆菌干预。连续喂养8周,留取小鼠粪便样本、尿液和血液,检测各组小鼠血糖、尿糖、24 h尿蛋白、血清肌酐、血清尿素氮水平。应用T-RFLP法分析小鼠肠道菌群。结果与模型组相比,双歧杆菌干预组双歧杆菌含量明显增加(P <0. 05),但仍小于正常对照组,肠道主要4种菌含量均无显著差异。双歧杆菌干预组血糖、尿糖、尿蛋白、血清肌酐、血清尿素氮水平比模型组低,差异具有统计学意义(P<0.05),但与正常对照组仍有差异。结论高糖饮食与STZ诱导小鼠可导致肠道菌群结构失调和肾功能受损,双歧杆菌干预可以改善高糖饮食与STZ诱导小鼠菌群失调以及肾功能。 Objective To explore the effect of bifidobacterium intervention on intestinal flora of mice with high glucose diet and STZ.Methods 30 female C57BL/6J mice were randomly divided into normal control group,model group and bifidobacterium intervention group,10 in each group,normal group was given standard feed,the model group was given high sugar feed and STZ injection,bifidobacterium intervention group was given high sugar feed,STZ injection and bifidobacterium intervention at the same time,8 weeks later,the fecal samples,urine and blood samples of mice were collected.Blood glucose,urine sugar,24 h urine protein,serum creatinine and serum urea nitrogen were measured.The intestinal flora of mice was analyzed by T-RFLP method.Results Compared with the model group,the content of bifidobacteria in bifidobacteria was significantly increased(P<0.05),but it was still less than the normal control group,and there was no significant difference in the contents of the main 4 bacteria in the intestinal tract.The levels of blood glucose,urine sugar,urine protein,serum creatinine and serum urea nitrogen were lower than those in the model group,and the differences were statistically significant(P<0.05),but they were still different from the control group.Conclusion hyperglycemic diet and STZ induced mice can lead to intestinal flora imbalance and impaired renal function,and bifidobacterium intervention can improve the diet and STZ induced microflora imbalance and renal function of mice.
