摘要
目的探讨lncRNA BLACAT1在不同WHO分级的星形细胞瘤样本的表达差异,以及BLACAT1对星形细胞瘤的生物学功能的影响。方法运用qRT-PCR技术检测本院星形细胞瘤样本的lncRNA BLACAT1表达量,分析lncRNA BLACAT1的表达量与WHO分级是否相关。运用RTCA增殖实验、划痕实验、Traswell侵袭/迁移实验检测NC组(阴性对照)和silence组(敲减BLACAT1)在细胞增殖、侵袭、迁移功能上的变化。结果星形细胞瘤WHO分级越高,lncRNA BLACAT1的表达量越高(P <0.001)。在RTCA增殖实验中,silence组增殖速率120h后明显慢于NC组(P <0.01),在划痕实验中,在12h、24h两个时间点测得silence组的划痕愈合速度慢于NC组(P <0.01)。Transwell侵袭、迁移实验中silence组24h后穿透到底面的细胞数量显著少于NC组(P <0.01)。结论星形细胞瘤分期分级越高,lncRNA BLACAT1的表达量越高。lncRNA BLACAT1对星形细胞瘤的增殖、侵袭、迁移能力有促进作用。
Objective We investigate the expression difference of long noncoding RNA of bladder cancer associated transcript 1(lncRNA BLACAT1)in different WHO grading astrocytoma specimens and the effects of lncRNA BLACAT 1 on the potential biological function of astrocytoma. Methods We used qRT-PCR technique to detect the expression of lncRNA BLACAT1 in clinical astrocytoma specimens from Yijishan Hospital of Wannan Medical College,and then analyzed whether the expression of lncRNA BLACAT1 was associated with WHO grading of astrocytoma.We detected the function changes of cell proliferation,invasion and migration in the negative control(NC)group and silence group(knockdown BLACAT1)by the application of Real Time Cell Analysis(RTCA)proliferation test,Scratch test,Transwell cell invasion/migration test. Results The higher the WHO grading and staging of astrocytoma was,the higher the lncRNA BLACAT1 expression was(P <0.001).RTCA test results showed that the proliferation rate of silence group was significantly slower than that of the negative control group after 120 hours(P <0.01).Scratch test results showed that the scratches of the silence group healed more slowly than those of the negative control group when measured at the time points of 12 hours and 24 hours(P <0.01). Conclusion of the Transwell migration and invasion assay showed that the number of astrocytoma cells penetrated to the bottom surface after 24 hours in the silence group was smaller than that in the NC group(P <0.01).Conclusion The higher the staging and grading of astrocytoma,the higher the expression of lncRNA BLACAT1.LncRNA BLACAT1 promotes the function of astrocytoma cell proliferation,invasion and migration.
