中风1号对急性脑出血大鼠神经功能和脑组织形态的影响

Effects of Zhongfeng No.1 on Nerve Function and Brain Tissue Morphology in Rats with Acute Cerebral Hemorrhage

摘要 目的观察中风1号汤剂对急性脑出血大鼠脑组织形态及神经功能的影响。方法将50只雄性SD大鼠随机分为5组(中风1号组、甘露醇组、模型组、假手术组、空白组),每组10只,通过自体注血方式造模并给药,评估中风1号对大鼠神经功能评分及脑组织含水量,同时取脑组织固定、脱水、包埋、固化、切片,应用透射电子显微镜观察中风1号对各组大鼠脑出血后血肿周围凋亡神经细胞超微体结构的影响。结果中风1号组术后5 d神经功能评分及脑组织含水量与模型组差异有统计学意义(P <0.01);电镜方面中风1号组较模型组核膜完整、清楚,其线粒体肿胀程度、细胞内细胞器数量等方面均优于模型组。结论中风1号汤剂能够减少急性脑出血大鼠脑组织的神经细胞凋亡,起到保护脑组织的作用。 Objective:To observe the effect of Zhongfeng No.1on the morphology of brain tissue in rats with acute cerebral hemorrhage,and to evaluate the effect of Zhongfeng No.1on the neurological function of rats by neurological function score and water content of brain tissue.Methods:50male SD rats were randomly divided into5groups(Zhongfeng No.1group,mannitol group,the model group,sham operation group and the blank group),10rats in each.Rats were modeled and administered by autologous blood injection.The neurologic function scores and brain tissue moisture contents were evaluated.The ultrastructure of apoptotic neurons around the hematoma of rats after intracerebral hemorrhage was observed by transmission electron microscopy.Results:There was a significant difference in neurologic function scores and brain tissue water content between Zhongfeng No.1group and the model group5days after surgery(P<0.01).Compared with the model group,the nuclear membrane in the Zhongfeng No.1group was intact and clear.The degree of mitochondrial swelling and the number of intracellular organelles were all better than those in the model group.Conclusion:Zhongfeng No.1can reduce the neuronal cell apoptosis in rats with acute cerebral hemorrhage and protect brain tissue.
