摘要
目的考察和厚朴酚对4T1细胞凋亡和肺转移模型生存期的影响。方法用不同浓度和厚朴酚处理肿瘤细胞(4T1、B16F10、H460、H1975)24、48、72 h,采用MTT法测定细胞半抑制率(IC50),采用Western blotting法测定细胞CleavedCaspase3蛋白的表达情况。同时建立Balb/c小鼠4T1细胞肺转移模型,考察和厚朴酚对小鼠生存期的影响。结果体外结果表明,和厚朴酚能明显抑制肿瘤细胞增殖,且随时间增加抑制作用相应增强,同时和厚朴酚可上调4T1细胞Cleaved-Caspase3蛋白表达,诱导细胞凋亡。体内结果表明,和厚朴酚可明显延长4T1细胞肺转移小鼠生存期。结论和厚朴酚能有效抑制肿瘤细胞增殖,其作用机制可能是通过上调凋亡相关蛋白Cleaved-Caspase3的表达来诱导细胞凋亡。同时,和厚朴酚能明显延长肿瘤肺转移模型小鼠的生存期。
Objective To investigate the impacts of honokiol on the apoptosis of breast cancer cell 4 T1 and the survival time of lung metastasis model of mice. Methods Cancer cells(4 T1, B16 F10, H460 and H1975) were treated with honokiol of different concentrations for 24, 48 and 72 hours; MTT was used to determine cell semi-inhibition rate(IC50) results, and Western blotting to determine the expression of Cleaved-Caspase3 protein in cells. Meanwhile, the lung metastasis model of cells 4 T1 in Balb/c mice was established to investigate the impacts of honokiol on the survival time of mice. Results In vitro results showed that honokiol can significantly inhibit the proliferation of cancer cells; and the inhibition increased with time. Besides, honokiol can promote the expression of Cleaved-Caspase3 protein in cells 4 T1 and induce the cells to apoptosis. In vitro results showed that honokiol can significantly prolong the survival time of 4 T1 lung metastatic mice. Conclusion Honokiol can effectively inhibit the proliferation of cancer cells. Its acting mechanism may be inducing cells to apoptosis by promoting the expression of Cleaved-Caspase3, an apoptosisrelated protein. In addition, honokiol can obviously prolong the survival time of mice with tumor lung metastasis model.
