癃畅颗粒对大鼠前列腺增生组织Caspase-3基因表达的影响 被引量:3

Effect of Longchang Granule on Expression of Caspase-3 Gene in Prostate Tissue of Rats with Benign Prostate Hyperplasia

摘要 目的:探讨癃畅颗粒治疗良性前列腺增生症(BPH)的作用机制。方法:将120只Wistar大鼠随机分为6组,即正常组、模型组、癃闭舒组、癃畅颗粒低剂量组、癃畅颗粒中剂量组、癃畅颗粒高剂量组,除正常组外,其余组大鼠采用去势后注射丙酸睾酮的方法建立BPH模型。造模的同时,正常组、模型组灌以等量生理盐水,余4个治疗组每天灌胃给药1次,连续30 d,采用原位细胞凋亡标记技术(TUNEL)检测前列腺组织细胞凋亡状况,免疫组织化学法检测前列腺组织细胞Caspase-3的表达状况。结果:模型组大鼠前列腺组织细胞凋亡指数(%)及Caspase-3表达百分率分别为(5.97±0.62)%、(16.00±1.46)%,癃闭舒组大鼠上述两指标分别为(81.43±6.40)%、(92.00±1.67)%,癃畅颗粒低剂量组两指标分别(25.46±3.97)%、(71.00±2.51)%,癃畅颗粒中剂量组分别为(46.50±3.56)%、(80.00±1.91)%,癃畅颗粒高剂量组分别为(81.20±7.66)%、(93.00±2.10)%;与模型组比较,癃闭舒组和癃畅颗粒各剂量组大鼠前列腺组织细胞凋亡指数及Caspase-3表达百分率均有不同程度升高(P<0.01);癃畅颗粒高剂量组与癃闭舒组比较无显著差异(P>0.05),但两组均显著高于癃畅颗粒中、低剂量组(P<0.01),癃畅颗粒中剂量组显著高于低剂量组(P<0.01)。结论:癃畅颗粒可以上调大鼠BPH组织中Caspase-3的表达,促进细胞凋亡,这可能是其治疗BPH的作用机制。 Objective To investigate the mechanism of Longchang Granule(LG)in the treatment of benign prostatic hyperplasia(BPH).Methods One hundred and twenty Wistar rats were randomly divided into6groups:normal group,model group,Longbishu group,low-dose,mid-dose and high-dose Longchang Granule groups.Except normal group,all the rest rats were castrated and injected with testosterone propionate to establish BPH models.All the model rats were killed after ig administered with Longchang Granule,Longbishu and normal saline respectively,once daily,for30d.The cell apoptosis was identified by TUNEL assay.IHC method was applied to detecting the expression of caspase-3gene in prostate tissue.Results The apoptotic index(%)and the percentage of caspase-3expression in the prostate tissue of the model group were(5.97±0.62)%and(16.00±1.46)%,the above two indexes of the Longbishu group were(81.43±6.40)%and(92.00±1.67)%,the two indicators of the low-dose Longchang Granule group were(25.46±3.97)%and(71.00±2.51)%,those in the mid-dose Longchang Granule group were(46.50±3.56)%and(80.00±1.91)%,respectively,and those in the high-dose Longchang Granule group were(81.20±7.66)%and(93.00±2.10)%,respectively.Compared with model group,the prostate apoptosis index of all treatment groups were significantly increased to different levels,also the expression of caspase-3gene increased obviously(P<0.01).No significant difference was found between high-dose Longchang Granule group and Longbishu group(P>0.05),however the two groups were significantly higher than those of the low-dose and mid-dose Longchang Granule groups(P<0.01).The mid-dose Longchang Granule droup significantly higher than the low-dose Longchang Granule group(P<0.01).Conclusion Longchang Granule can upregulate the expression of caspase-3gene in prostate tissue of BPH rats and accelerate cell poptosis.This is the possible mechanism of Longchang Granule in the treatment of BPH.
