耳针对氧化偶氮甲烷致直肠癌大鼠食欲的影响及其机制研究

Effect and mechanism of auricular acupuncture on appetite in the rats AOM-Induced Rectal Cancer

目的探讨耳针对氧化偶氮甲烷(AOM)致直肠癌大鼠食欲的影响及其机制研究。方法用AOM腹腔注射诱导大鼠发生直肠癌,从造模开始,每周对耳针组、体针组大鼠进行针刺治疗,观察耳针对大鼠体重、摄食量、血清食欲素A、瘦素、胸腺指数、脾脏指数以及直肠病理组织学的影响。结果用AOM成功诱导了大鼠直肠癌模型,正常对照组未见肿瘤发生,模型组、耳针组、体针组各组大鼠直肠部分均不同程度出现肿瘤;与模型组比较,第10、15、20周体针组、耳针组体重明显增加,其中第15周体针组、耳针组体重显著增加(P <0. 05);与模型组比较,第5、10、15、20周体针组、耳针组大鼠摄食量明显增加,其中第5周体针组、耳针组摄食量显著增加(P <0. 05,P <0. 01);与正常组比较,模型组大鼠血清食欲素A、瘦素均显著升高(P <0. 01);与模型组比较,耳针组、体针组大鼠血清食欲素A、瘦素均显著降低(P <0. 01);与正常组比较,模型组、耳针组、体针组脾脏指数均有升高趋势,胸腺指数均有下降趋势;但各组大鼠脾脏指数、胸腺指数变化差异均无统计学意义。结论耳针治疗可以明显增强AOM致直肠癌大鼠摄食量,可能与耳针刺激使得血清中廋素、食欲素A含量变化有关。

Objective To explore the effect and mechanism of auricular acupuncture on appetite in the rats of rectal cancer induced by azoxymethane(AOM).Methods The intraperitoneal injection of AOM was used to induce rectal cancer in rats.Since modeling,acupuncture was given in the auricular acupuncture group and the body acupuncture group.The body mass,food intake,serum orexin A,leptin,thymus index,spleen index and the rectal historical effects were ob-served in the treatment with auricular acupuncture.Results In the research,the rectal cancer models were successfully in-duced with AOM in the rats.No tumor was found in the normal control group.Tumors were discovered in the rectum of the rats in each of the model group,the auricular acupuncture group and the body acupuncture group.Compared with the model group,in the week10,15and20,the body masses were increased obviously in the body acupuncture group and the auricu-lar acupuncture group,in which,the body mass was increased significantly in the week15(P<0.05).Compared with the model group,in the week10,15and20,the food intake was increased obviously in the body acupuncture group and the au-ricular acupuncture group,in which,in the week5,the food intake was increased significantly(P<0.05,P<0.01).Com-pared with the normal group,serum orexin A and leptin were increased significantly in the model group(P<0.01).Com-pared with the model group,they were reduced significantly in the body acupuncture group and the auricular acupuncture group(P<0.01).Compared with the normal group,the spleen indexes were all increased in tendency in the model group,the body acupuncture group and the auricular acupuncture group and the thymus indexes were all reduced in tendency.The differences were not significant in spleen index and thymus index.Conclusion The auricular acupuncture remarkably in-creases the food intake in the rats of AOM-induced rectal cancer,which is probably related to the changes of serum leptin and orexin A after the stimulation with auricular acupuncture.
