坤灵丸对子宫内膜增生模型大鼠的实验研究

Experimental Study of Kunling Pill on Rats with Endometrial Hyperplasia

目的:研究分析坤灵丸对肾虚血瘀型子宫内膜增生症大鼠子宫内膜形态学及血清六项激素的影响。方法:选取SPF级雌性SD大鼠70只,将其随机分为空白组10只,造模组60只。造模参照《雌性大鼠肾虚血瘀模型初探》建立肾虚血瘀型子宫内膜增生大鼠模型。随后将模型组60只大鼠随机分为模型组、坤灵丸组、补肾活血方组、米非司酮组共4组,每组15只,分别灌服相应药液。给药21 d后处死大鼠,剖开腹腔,取出子宫,HE染色观察子宫形态学变化。腹主动脉取血,分离血清,检测六项激素含量。结果:与空白组比较,模型组子宫内膜厚度增加;与模型组比较,各用药组子宫内膜厚度减少,子宫内膜腺体数量多,腺体弯曲,腺上皮多呈单层高柱状上皮细胞。与空白组比较,模型组大鼠血清雌激素(E2)、泌乳素(Prol)、黄体生成素(LH)、卵泡雌激素(FSH)含量均明显升高(P <0. 01),孕激素(P)、睾酮(T)含量明显降低(P <0. 01);与模型组比较,各用药组大鼠血清E2、Prol、LH、FSH含量均明显降低(P <0. 01或P <0. 05),P、T含量明显升高(P <0. 01或P <0. 05)。结论:坤灵丸和补肾活血方能够降低子宫内膜厚度,改善血清激素水平。

Objective:To study and analyze the effects of Kunling Pill on endometrial morphology and serum six hormones in rats with endometrial hyperplasia of kidney deficiency and blood stasis type.Methods:Seventy SPF female SD rats were randomly divided into blank group(10rats)and model group(60rats).The rat model of endometrial hyperplasia of kidney deficiency and blood stasis type was established hy reierring to Preliminary Study on Female Rat Model of Kidney Deficiency and Blood Stasis.Subsequently,60rats in the model group were randomly divided into four groups:model group,Kunling pill group,Bushen Huoxue recipe group and Mifepristone group,and15rats in each group were given corresponding medicines respectively.After21days of administration,rats were sacrificed,abdominal cavity was opened,uterus was taken out,and uterus morphological changes were observed by HE staining.Blood was taken from abdominal aorta,serum was separated and six hormones were detected.Results:Compared with the blank group,the thickness of endometrium in the model group increased;compared with the model group,the thickness of endometrium in the treated groups decreased,the number of endometrial glands was large,the glands were curved,and the glandular epithelium was mostly monolayer high columnar epithelial cells.Compared with the blank group,the serum levels of estrogen(E2),prolactin(Prol),luteinizing hormone(LH),follicular estrogen(FSH)in the model group increased significantly(P<0.01),while the levels of progesterone(P)and testosterone(T)decreased significantly(P<0.01);compared with the model group,E2,Prol,LH and FSH in the treated groups were significantly decreased(P<0.01or P<0.05),while the content of P and T increased significantly(P<0.01or P<0.05).Conclusion:Kunling Pill and Bushen Huoxue Prescription can reduce the thickness of endometrium and improve the level of serum hormones.
