Identification of T-cell epitopes of major peach allergen Pru p 3

摘要 目的 Pru p 3是桃主要致敏蛋白,目前尚未见桃Pru p 3 T细胞表位在我国桃过敏反应中的研究,本研究探讨桃主要致敏蛋白Pru p 3 T细胞表位。方法纳入蒿花粉相关桃过敏患者20例和健康对照者10人,采集受检者的外周血单个核细胞(PBMC),将PBMC分别与8条Pru p 3来源的肽段共同孵育,比较桃过敏患者和健康对照者PBMC受各肽段刺激后的增生水平,分析不同肽段的变应原性。结果肽段Pru p 367-82对桃过敏患者PBMC的活化程度最高,20例桃过敏患者中对该肽段刺激指数(SI)阳性者14例,占70%; Pru p 323-38对桃过敏患者PBMC的活化率为55%(11/20)。Pru p 367-82与Art v 390-105氨基酸序列的一致率为81%。结论 Pru p 323-38和Pru p 367-82是引起我国桃过敏反应的主要T细胞表位,Pru p 367-82可能是参与蒿花粉-桃交叉反应的T细胞表位。 Objective Pru p 3 is the major allergen of peach.T-cell epitopes of Pru p 3 have not been investigated among Chinese peach allergic patients yet.The objective of this study was to map the major T-cell epitopes of Pru p 3.Methods Peripheral blood mononuclear cells(PBMCs)from 20 peach-allergic patients were cultured with a panel of 8 synthetic peptides overlapping for 5 aa residues and representing the full sequence of Pru p 3.Proliferation in 5-day assays was carried out via CCK-8 incorporation.The results were compared to 10 non-peach allergic volunteers.Results Highest activation capacity was shown by two peptides.Activation rate of PBMCs was 70%and 55%which was induced by Pru p 3 67-82 and Pru p 3 23-38 among peach-allergic patients respectively.Concordance rate of sequence was 81%between mugwort pollen allergen Art v 3 90-105 and peach allergen Pru p 3 67-82.Conclusions Two immunodominant T-cell-reactive regions of Pru p 3 were identified:Pru p 3 23-38 and Pru p 3 67-82.Pru p 3 67-82 might be the relevant epitope for cross-reacting with Artemisia pollen.
