回医烙灸疗法对大鼠颈椎间盘退变模型软骨细胞凋亡的影响

Effect of Moxibustion in Hui Medicine on Chondrocyte Apoptosis in Degenerated Cervical Intervertebral Discs of Rats Models

目的探讨回医烙灸疗法对颈椎间盘退变模型治疗作用及其机制。方法选取30只3月龄雌性SD大鼠随机分为空白对照组、模型对照组和烙灸组,每组10只,采用切断右侧颈部肌群的方法制成动静力失衡型颈椎间盘软骨退变模型,烙灸组在造模4周确认成功后,进行烙灸疗法,每日1次,每次20min,干预治疗4周。空白对照组和模型对照组不予干预。烙灸治疗4周后,行颈椎正侧位X线片评分及软骨细胞凋亡指数检测。结果颈椎影像学评分方面:烙灸组[(2.02±0.18)分]与空白对照组[(0.58±0.13)分]低于模型对照组[(5.08±1.59)分]评分(P<0.05),烙灸组与空白对照组比较差异无统计学意义(P>0.05)。颈椎间盘软骨细胞凋亡指数(荧光染色)烙灸组(20.13±3.51)和空白对照组(6.13±0.72)低于模型对照组(40.23±2.13)(P<0.05);烙灸组间盘终板软骨细胞凋亡指数高于空白对照组(P<0.05)。结论回医烙灸疗法可减少大鼠颈椎间盘退变模型终板软骨细胞的凋亡,从而延缓颈椎间盘软骨的退变。

Objective To explore the therapeutic effect and mechanism of moxibustion therapy on cervical disc degeneration model.Methods Thirty female SD rats,three month old,were randomly divided into three groups:blank group,model group and moxibustion group,with ten rats in each group.Dynamic-Static imbalance and Degenerated Cervical Intervertebral Disc rat model was made by cutting off the right neck muscles.After 4 weeks of model establishment,the moxibustion group received moxibustion therapy once a day for 20 minutes each time for 4 weeks.The blank control group and model group were not intervened.After 4 weeks of moxibustion treatment,the positive and lateral X-ray scores of cervical spine and the apoptotic index of chondrocyte were measured.Results The cervical imaging scores of moxibustion group(2.02±0.18)score and blank group(0.58±0.13)score were significantly lower than those of model group(5.08±1.59)score(P<0.05).The difference between moxibustion group and blank group was not statistically significant(P>0.05).The chondrocyte apoptosis indexes of the endplate cartilage cells in the moxibustion group(20.13±3.51)and the blank group(6.13±0.72)were significantly lower than that in the model group(40.23±2.13)(P<0.05),and moxibustion group was higher than blank group(P<0.05).Conclusion Moxibustion in Hui medicine can improve the local circulation of the cervical spine in rats,and reduce the apoptosis of endplate chondrocytes in the rat cervical disc degeneration model so to delay the degeneration of cervical intervertebral disc cartilage.
