摘要
[目的]对玉米南方锈病性状进行遗传分析,筛选南方锈病基因连锁的SSR引物标记,并进行初步定位。[方法]以抗南方锈病玉米自交系K381和感病自交系LX9801为材料,构建6个世代群体(P_1、P_2、F_1、F_2、B_1CF_1、B_2CF_1),对玉米南方锈病进行遗传分析,利用SSR标记对南方锈病连锁基因进行引物筛选,并利用筛选的分子标记对辅助抗南方锈病材料进行前景选择。[结果] F_2代分离群体抗病株与感病株比例符合3∶1分离比例,B_2CF_1[(K381×LX9801)×LX9801]群体抗感株接近1∶1分离比例,推测其抗性基因为单一显性基因控制的,与10.02区域内的2对SSR引物phi059和umc2018紧密连锁,初步认为K381的抗南方锈病基因位于第10号染色体短臂上。[结论]引物phi059可以用于玉米抗南方锈病材料辅助选择。
[Objective]To study the genetic analysis and SSR link marker screening of gene resistance to southern rust.[Method]Using K381(immune inbred line),LX9801(susceptible inbred line)and the derived population P1,P2,F1,F2,B1CF1and B2CF1as materials,we investigated the heredity of disease resistance gene and selected SSR marker to maize southern rust.SSR marker was used to assist foreground selection.[Result]The proportion of resistant and susceptible strains in F2population accorded with3∶1isolation ratio.And the ratio of B2CF1was1∶1.It was speculated that the resistance gene was controlled by a single dominant gene,closely linked with2pairs of SSR primers phi059and umc2018in region10.02.It was preliminarily believed that the anti-rust gene of K381located on the short side of chromosome10.[Conclusion]The primer phi059can be used to assist the selection of resistance to southern rust materials.
